Myelin Basic Protein Antibody / CNS Myelin Marker detects myelin basic protein (MBP), a major structural component of the myelin sheath encoded by the MBP gene. MBP is highly expressed by oligodendrocytes in the central nervous system (CNS), where it plays an important role in the formation, compaction and stability of myelin. By supporting the specialized membrane structure that surrounds axons, MBP contributes to efficient nerve impulse conduction and normal nervous system function.

MBP is a highly basic protein concentrated primarily on the cytoplasmic face of compact myelin membranes. Its interactions with membrane lipids help bring adjacent membrane layers together and maintain the tightly packed organization of the myelin sheath. Alternative splicing of the MBP gene produces multiple protein isoforms that vary in molecular weight and show developmental differences in expression. These properties have made MBP an established CNS myelin marker for investigating oligodendrocyte differentiation, myelin formation and maturation.

Changes in MBP expression, localization and integrity are studied in conditions involving myelin loss or damage. MBP is particularly relevant to multiple sclerosis and experimental autoimmune encephalomyelitis, where it can function as a myelin-associated autoantigen. MBP expression is also examined in studies of demyelination and remyelination, traumatic injury, neurodegeneration and nervous system development. Detection of this CNS myelin marker can therefore provide information about myelin distribution and changes associated with oligodendrocyte and white matter biology.

Clone MBP/4277R is a recombinant rabbit monoclonal antibody validated for MBP detection by immunohistochemistry and Western blot using human brain samples. Recombinant production provides a defined antibody sequence and supports manufacturing consistency. The antibody has also been evaluated on a HuProt protein microarray containing more than 19,000 full-length human proteins. MBP produced the dominant signal with a Z-score of 42.24 and an S-score of 22.43. The Z-score reflects signal intensity relative to proteins across the array, while the S-score reflects separation of the intended target from other detected proteins, providing additional evidence of target specificity.
