Mesothelin (MSLN) is a glycosylphosphatidylinositol-anchored cell surface protein with highly restricted expression in normal mesothelial cells but marked overexpression in ovarian carcinoma, particularly high-grade serous ovarian cancer. Mesothelin Antibody is widely used as an ovarian cancer marker antibody due to its consistent and often diffuse expression in tumor cells and its established role in tumor progression. Its interaction with MUC16 (CA125) promotes cell adhesion and facilitates peritoneal dissemination, a defining feature of advanced ovarian cancer, making mesothelin a biologically and clinically relevant biomarker.
Mesothelin antibody, also referred to as MSLN antibody or ovarian tumor antigen antibody, produces characteristic membranous and cytoplasmic staining corresponding to cell surface localization and protein processing. In tissue-based assays, normal expression is largely confined to mesothelial linings, while ovarian carcinoma demonstrates strong and uniform staining across malignant epithelial cells. This pronounced contrast between limited physiologic expression and tumor-associated overexpression enhances interpretability and supports reliable identification of ovarian tumor cells.
As an ovarian cancer marker antibody, this clone provides robust detection of ovarian serous carcinoma, where prominent membranous staining highlights tumor architecture and supports evaluation of tumor distribution within tissue sections. Mesothelin expression is also observed in other malignancies such as mesothelioma and pancreatic adenocarcinoma; however, its strong and consistent expression in ovarian cancer, combined with minimal staining in many non-expressing tissues, reinforces its value in tumor classification and biomarker analysis.
This antibody is further supported by western blot validation in OVCAR-3 ovarian cancer cell lysates, where it detects a band consistent with endogenous mesothelin expression. This confirms target recognition at the protein level and provides complementary evidence supporting its use across multiple experimental approaches, including immunohistochemistry and protein analysis workflows.
Functionally, mesothelin contributes to tumor cell adhesion, invasion, and metastatic spread through binding to MUC16 (CA125), promoting tumor implantation within the peritoneal cavity.
