MBP Antibody / Oligodendrocyte Marker Antibody detects myelin basic protein (MBP), a major structural protein of the myelin sheath produced by oligodendrocytes in the central nervous system (CNS). Encoded by the MBP gene, myelin basic protein contributes to the formation and maintenance of compact myelin surrounding neuronal axons. MBP is abundant in CNS myelin and is widely studied in neuroscience as a marker of oligodendrocytes, myelination and myelin integrity.
MBP is a highly basic protein that associates with the cytoplasmic surface of myelin membranes. Its interactions with negatively charged membrane lipids help bring adjacent membrane layers together, contributing to the tightly compacted structure of mature myelin. Alternative splicing of the MBP gene generates multiple isoforms with differences in molecular weight and developmental expression. These isoforms are associated with different stages of oligodendrocyte development and myelin maturation.
As an oligodendrocyte marker, MBP is commonly investigated in studies of glial cell differentiation, CNS development, myelin formation and axon-glia interactions. Changes in MBP expression or integrity are also associated with myelin damage and are studied in experimental models of demyelination and remyelination. MBP is particularly relevant to multiple sclerosis and experimental autoimmune encephalomyelitis, where myelin proteins can become targets of immune responses.
Clone rMBP/4288 is a recombinant mouse monoclonal antibody generated against a region within amino acids 150-250 of human MBP. The antibody has been validated by immunohistochemistry using FFPE human cerebellum and by Western blot using human brain lysate, where MBP isoforms may be detected from approximately 20-37 kDa. These data support detection of MBP in human neural tissue and lysates.
