Involucrin Antibody IVRN/2113R recognizes involucrin, a structural protein encoded by the IVL gene and expressed during terminal differentiation of stratified squamous epithelial cells. Involucrin is a major component of the cornified cell envelope and is widely used as a marker of keratinocyte differentiation. Its expression increases as keratinocytes leave the proliferative basal compartment and progress through the suprabasal layers toward terminal differentiation.
Involucrin is a soluble cytoplasmic protein before its incorporation into the developing cornified envelope. The human protein contains an unusual central region rich in tandem repeats and glutamine residues. During terminal differentiation, transglutaminases catalyze covalent crosslinking of involucrin to other structural proteins and lipids near the inner surface of the plasma membrane. This process contributes to formation of the highly insoluble protein-lipid structure that ultimately replaces the conventional plasma membrane in mature corneocytes.
The cornified envelope provides an essential mechanical and permeability barrier in the epidermis and other stratified squamous epithelia. Involucrin participates in the early stages of envelope assembly and provides a substrate for subsequent crosslinking events involving additional differentiation-associated proteins. Its regulated expression and incorporation into this structure make IVL an important marker for studying epidermal maturation, epithelial differentiation, and cornification.
Involucrin has a calculated molecular weight of approximately 68 kDa but characteristically migrates at a substantially higher apparent molecular weight during SDS-PAGE. This anomalous electrophoretic behavior is associated with the protein's unusual glutamine-rich, repetitive amino acid composition and should be considered when interpreting involucrin western blot results.
Changes in involucrin expression have been investigated in disorders of epidermal differentiation and in squamous epithelial neoplasia. Because IVL expression is closely associated with differentiation state, involucrin is also frequently examined in experimental systems studying keratinocyte maturation, epithelial development, and mechanisms controlling the transition from proliferating basal cells to differentiated suprabasal cells.
An Involucrin Antibody such as clone IVRN/2113R provides a recombinant monoclonal reagent for investigating involucrin expression and epithelial differentiation, while an IVL Monoclonal Antibody provides gene-based terminology for studies of keratinocyte maturation and cornified envelope formation.

1. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
Involucrin purified from human keratinocytes was used as the immunogen for the Involucrin Antibody IVRN/2113R.
Store the recombinant Involucrin Antibody IVRN/2113R at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
