BRAF N-Terminal Antibody RM308 Detection in K562 Leukemia Cells WB. Western blot analysis of human K562 chronic myelogenous leukemia cell lysate using BRAF N-Terminal Antibody clone RM308 at 1:100 dilution. A distinct immunoreactive band is detected at approximately 84 kDa, consistent with the expected molecular weight of full-length BRAF.
BRAF N-Terminal Antibody RM308 Staining of Human Kidney IHC. Immunohistochemistry staining of FFPE human kidney tissue using BRAF N-Terminal Antibody clone RM308 at 1:500 dilution. Heat-induced epitope retrieval was performed by steaming sections in pH 6 citrate buffer for 20 min followed by cooling. BRAF immunoreactivity is prominent in renal tubular epithelial cells, with predominantly cytoplasmic staining, while glomerular structures show comparatively limited staining.

Description

BRAF N-Terminal Antibody RM308 / BRAF N-Terminus Antibody detects the N-terminal region of BRAF, a serine/threonine protein kinase and member of the RAF family. BRAF functions within the RAS-RAF-MEK-ERK signaling pathway, transmitting signals from activated RAS proteins to MEK and ERK. This pathway regulates cellular proliferation, differentiation, survival and responses to extracellular growth signals.

The BRAF protein contains an N-terminal regulatory region and a C-terminal serine/threonine kinase domain. The N-terminal portion includes conserved regulatory elements involved in controlling BRAF activation and interactions with upstream signaling components. Recognition of this region differentiates a BRAF N-Terminus Antibody from reagents directed against other portions of the protein and can be useful when antibody epitope location is an important consideration in experimental design.

BRAF is extensively studied in cancer because activating alterations can produce sustained MAPK pathway signaling. The V600E substitution within the kinase domain is the best-known BRAF mutation and occurs in melanoma as well as thyroid, colorectal, lung and other cancers. Other BRAF alterations, including mutations, fusions and changes affecting regulatory regions, further illustrate the diverse mechanisms through which abnormal BRAF signaling can contribute to disease.

Clone RM308 is a recombinant rabbit monoclonal antibody generated against a peptide corresponding to residues near the N-terminus of human BRAF. Its defined N-terminal targeting provides a differentiated reagent for researchers examining BRAF expression and RAF-MAPK pathway biology.
