PRKCI Antibody / Atypical Protein Kinase C Antibody recognizes protein kinase C iota (PRKCI), a calcium- and diacylglycerol-independent serine/threonine kinase belonging to the atypical protein kinase C subfamily. Unlike conventional and novel PKC isoforms, PRKCI is activated primarily through protein-protein interactions and phosphorylation events, allowing it to regulate diverse signaling pathways involved in cell polarity, proliferation, migration, differentiation, and survival. PRKCI is expressed in numerous epithelial, neural, and immune tissues, where it contributes to the establishment and maintenance of normal cellular architecture and signaling.
PRKCI is a core component of the PAR polarity complex, functioning alongside PAR3 and PAR6 to establish apical-basal polarity and regulate asymmetric cell division. These conserved functions are essential during embryonic development and remain important in adult tissues requiring continuous renewal or regeneration. Beyond its role in cell polarity, PRKCI integrates signaling downstream of phosphoinositide 3-kinase, AKT, small GTPases, and additional regulatory pathways that influence cell growth and survival. PRKCI also contributes to regulation of the NF-kappa B pathway, apoptosis resistance, and directed cell migration. Aberrant PRKCI expression or activation has been reported in numerous malignancies, including cancers of the lung, ovary, pancreas, colon, and brain, where increased activity has been associated with tumor progression, invasion, and therapeutic resistance. Consequently, PRKCI has emerged as both a valuable biomarker and a potential therapeutic target in oncology research.
