PPIG Antibody by WB in Human, Rat, and Mouse Cell Lysates. Western blot analysis of Cyclophilin G in human K562 (lane 1), HEL (lane 2), HepG2 (lane 3), and RT4 (lane 4) cells; rat RH35 (lane 5) and C6 (lane 6) cells; and mouse Hepa1-6 (lane 7) and Neuro-2a (lane 8) cells. Samples containing 30 ug of protein per lane were separated by 10% SDS-PAGE under reducing conditions. The rabbit polyclonal antibody was used at 0.5 ug/ml and detected a prominent band at approximately 120 kDa across all eight samples, compared with the calculated molecular weight of 88.6 kDa. This higher apparent molecular weight is consistent with independent reports of PPIG migration near 110 kDa and may reflect anomalous electrophoretic mobility associated with its serine/arginine-rich regions and phosphorylation. Additional lower-molecular-weight bands are visible in several lanes.

Description

PPIG Antibody recognizes peptidyl-prolyl cis-trans isomerase G (PPIG), a member of the cyclophilin family involved in protein folding, RNA processing, and regulation of gene expression. Also known as Cyclophilin G, this protein functions as a peptidyl-prolyl isomerase (PPIase), an enzyme that catalyzes the interconversion of cis and trans peptide bonds preceding proline residues. This activity can influence protein conformation, molecular interactions, and the assembly of protein complexes involved in essential cellular processes.

Unlike smaller cyclophilin family members, PPIG contains an extended C-terminal region enriched in serine-arginine (SR) dipeptide motifs. These structural features distinguish Cyclophilin G from other peptidyl-prolyl isomerases and support its association with proteins involved in precursor messenger RNA (pre-mRNA) processing. PPIG combines a conserved cyclophilin catalytic domain with additional interaction regions that contribute to its specialized functions within the nucleus.

A major biological role of PPIG involves its association with the spliceosome, a large ribonucleoprotein complex responsible for removing introns from pre-mRNA. Spliceosome assembly and activity require coordinated interactions among small nuclear ribonucleoproteins, splicing factors, and regulatory proteins. Cyclophilin G has been identified in spliceosomal complexes, where its protein interaction capabilities may contribute to the organization and conformational regulation of RNA-processing machinery.

PPIG is also associated with nuclear speckles, subnuclear compartments enriched in splicing factors and other RNA-binding proteins. These structures help organize components involved in messenger RNA maturation and alternative splicing. Through its interactions with spliceosomal proteins, PPIG may influence the dynamics of RNA-processing complexes and the regulation of transcript maturation.

Alternative splicing allows individual genes to produce multiple transcript variants, contributing to protein diversity and cellular specialization. Because PPIG participates in protein interactions associated with RNA processing, it is relevant to studies of gene expression, nuclear organization, and post-transcriptional regulation. Understanding Cyclophilin G function may also provide insight into how peptidyl-prolyl isomerization contributes to the assembly and activity of complex molecular machinery.

A PPIG Antibody from NSJ Bioreagents supports research investigating Cyclophilin G, spliceosomal protein interactions, RNA metabolism, and the molecular mechanisms regulating pre-mRNA splicing.
