Phospho-YAP1 (Ser127) Antibody recognizes Yes Associated Protein 1 phosphorylated at serine 127, a critical regulatory site controlling YAP1 activity and subcellular localization. YAP1 is a transcriptional coactivator and major downstream effector of the Hippo signaling pathway. In response to activation of Hippo kinases, large tumor suppressor kinases phosphorylate YAP1 at Ser127, creating a binding site for 14-3-3 proteins that promotes cytoplasmic retention and prevents nuclear accumulation. This phosphorylation event inhibits YAP1 mediated transcription and serves as a key mechanism for restricting cell proliferation and maintaining tissue homeostasis.
YAP1 integrates signals from cell density, extracellular matrix stiffness, mechanical stress, and growth factor pathways to regulate organ size, differentiation, and tissue regeneration. The balance between phosphorylated and unphosphorylated YAP1 determines whether cells maintain a quiescent state or initiate growth promoting transcriptional programs. Reduced phosphorylation at Ser127 permits nuclear localization and activation of TEAD transcription factors, resulting in expression of genes involved in proliferation, survival, stemness, and epithelial mesenchymal transition. Consequently, phosphorylation of YAP1 at Ser127 serves as an important indicator of Hippo pathway activity and cellular growth control.
Abnormal regulation of YAP1 phosphorylation has been implicated in numerous diseases, including hepatocellular carcinoma, pancreatic cancer, lung cancer, breast cancer, ovarian cancer, colorectal cancer, and glioblastoma. Decreased Ser127 phosphorylation and increased nuclear YAP1 activity have been associated with tumor progression, metastasis, cancer stem cell maintenance, and resistance to chemotherapy. In addition to oncology, altered YAP1 signaling contributes to fibrosis, cardiovascular disease, and regenerative responses following tissue injury. Measurement of phospho YAP1 provides valuable information regarding pathway activation and therapeutic responses.
