Phospho-PTEN (pY336) Antibody / Phospho-PTEN Tyr336 Antibody recognizes PTEN when phosphorylated at tyrosine 336. PTEN, or phosphatase and tensin homolog, is a major tumor suppressor that negatively regulates phosphoinositide 3-kinase (PI3K)/AKT signaling. Through its lipid phosphatase activity, PTEN reduces levels of phosphatidylinositol (3,4,5)-trisphosphate (PIP3), thereby limiting downstream signals involved in cellular proliferation, survival, growth and metabolism.

PTEN function is regulated through multiple post-translational mechanisms, including phosphorylation at serine, threonine and tyrosine residues. Tyr336 is located near the C-terminal portion of the PTEN C2 domain, a region involved in membrane association and interactions that contribute to PTEN localization and regulation. A PTEN (pY336) Antibody enables researchers to examine this site-specific phosphorylated form separately from total PTEN protein.

Phosphorylation at Tyr336 has been studied in relation to the regulation of PTEN protein stability. Tyrosine phosphorylation can alter interactions between PTEN and regulatory proteins, potentially affecting its susceptibility to degradation and the amount of functional PTEN available within the cell. Because these regulatory changes can occur without corresponding changes in PTEN gene expression, analysis of Tyr336 phosphorylation can provide information that complements measurements of total PTEN abundance.

Regulation of PTEN stability and activity is particularly important in cancer biology. Reduced PTEN function can permit increased PI3K/AKT pathway signaling and contribute to abnormal cellular proliferation and survival. PTEN is frequently disrupted in human tumors through genetic alterations, reduced expression and changes in post-translational regulation. Phospho-specific analysis provides a way to investigate regulatory states of PTEN that may influence tumor suppressor activity even when the protein remains expressed.
