Pan-HLA Class II Antibody HLA-Pan/8311R / HLA-DP/DQ/DR Antibody recognizes the classical human MHC class II molecules HLA-DP, HLA-DQ and HLA-DR. MHC class II proteins provide a molecular interface between antigen-presenting cells and CD4-positive T lymphocytes. By displaying peptides generated from internalized proteins, these cell-surface complexes allow helper T cells to recognize antigen and participate in the regulation of downstream cellular and humoral immune responses.

Each classical HLA class II molecule consists of an alpha and beta chain that associate non-covalently at the cell surface. Both subunits contribute to an open-ended peptide-binding groove that accommodates processed antigenic peptides. Newly synthesized class II molecules are directed through intracellular compartments associated with antigen processing, where peptides can be loaded before the resulting complexes are transported to the plasma membrane for T cell recognition.

HLA-DP, HLA-DQ and HLA-DR are encoded by distinct loci within the human MHC and exhibit substantial genetic polymorphism. Variation within these molecules influences peptide-binding preferences and contributes to differences in antigen recognition among individuals. This diversity has made MHC class II biology particularly important in research involving autoimmune susceptibility, infectious disease, transplantation compatibility and immune responses to tumors.

Pan-HLA class II detection provides a complementary strategy to analysis with locus-specific HLA antibodies. Rather than distinguishing DP, DQ or DR individually, broad recognition can be used to investigate the overall distribution and regulation of MHC class II molecules. Changes in HLA class II expression may accompany immune-cell differentiation, activation and inflammatory signaling, providing additional context for studies of antigen presentation and immune regulation.
