Description

p73 is a member of the p53 family of transcription factors and plays a vital role in regulating cell cycle arrest, apoptosis, DNA repair, and neuronal development. Although structurally and functionally related to p53, p73 has distinct regulatory pathways and biological roles. It is activated in response to cellular stress and contributes to tumor suppression, but unlike p53, it is rarely mutated in human cancers.

The p73 protein is encoded by the TP73 gene, which undergoes complex alternative splicing, resulting in multiple isoforms with varying transcriptional activities. Two major classes of isoforms exist: the transactivation-competent TAp73 isoforms that promote apoptosis and the N-terminal truncated ΔNp73 isoforms that can act as dominant-negative inhibitors of both p53 and TAp73. The balance between these isoforms can significantly influence cellular responses to stress and the development of malignancies.
