P15RS Antibody Human A549 Lung Cancer Cell IF. Immunofluorescent staining of human A549 cells was performed with P15RS Antibody at 5 ug/ml and Alpha Tubulin antibody overnight at 4oC. Enzyme antigen retrieval was performed for 15 minutes. P15RS/RPRD1A was detected with Fluoro488-conjugated secondary antibody (green), while Alpha Tubulin was detected with Cy3-conjugated secondary antibody (red). P15RS staining is observed predominantly in the nuclear and perinuclear regions, while Alpha Tubulin highlights the cytoskeletal network. This P15RS Antibody demonstrates localization of RPRD1A in human A549 lung cancer cells.

P15RS Antibody Human, Mouse and Rat WB. Western blot analysis was performed with P15RS Antibody on human 293T, A431, Jurkat and SH-SY5Y cell lysates (lanes 1-4), rat brain and C6 cell lysates (lanes 5-6), and mouse brain tissue lysate (lane 7). Samples (30 ug) were separated by 10% SDS-PAGE under reducing conditions, and the antibody was used at 0.25 ug/ml overnight at 4oC. A prominent band was detected at approximately 36 kDa, consistent with the expected molecular weight of RPRD1A. P15RS Antibody Human SH-SY5Y Neuroblastoma Cell IP. Immunoprecipitation of P15RS/RPRD1A from human SH-SY5Y neuroblastoma whole cell lysate was performed with P15RS Antibody. Input lysate (lane 1), rabbit control IgG IP (lane 2) and P15RS Antibody IP (lane 3) were analyzed by western blot. A specific RPRD1A band is detected at approximately 36 kDa in the input and antibody immunoprecipitate, consistent with the expected molecular weight. The prominent approximately 50 kDa signal represents IgG heavy chain. This P15RS Antibody demonstrates immunoprecipitation of RPRD1A from human SH-SY5Y neuroblastoma cells.

Description

P15RS Antibody / RPRD1A Antibody detects RPRD1A, a nuclear protein involved in transcriptional regulation and RNA polymerase II-associated processes. RPRD1A, historically known as p15RS, contains a CTD-interacting domain that enables association with the C-terminal domain of RNA polymerase II. Through these interactions, RPRD1A participates in mechanisms coordinating transcription and RNA processing, making it a useful target for studies of gene expression.

RNA polymerase II contains a C-terminal domain that undergoes dynamic phosphorylation during different stages of transcription. These phosphorylation patterns regulate the recruitment of proteins involved in transcription initiation, elongation, termination and RNA processing. RPRD family proteins interact with the phosphorylated RNA polymerase II CTD and have been associated with regulation of CTD phosphorylation through interactions with CTD phosphatases. A P15RS Antibody can therefore support investigation of mechanisms controlling RNA polymerase II activity and transcriptional progression.

Beyond its association with transcriptional machinery, p15RS has been investigated in pathways controlling cell proliferation and differentiation. RPRD1A has been linked to Wnt signaling and beta-catenin-dependent transcription, an important regulatory pathway in development, tissue homeostasis and cancer biology. These findings connect RPRD1A with both nuclear transcriptional control and signaling mechanisms that influence cellular growth.

Dysregulation of transcription and proliferation is a common feature of cancer. RPRD1A expression and activity are therefore of interest in studies examining tumor biology, gene-expression control and signaling pathways associated with abnormal cell growth. Investigation of p15RS may also help clarify how transcription-associated proteins integrate growth-regulatory signals with RNA polymerase II function.
