ErbB2 receptor tyrosine kinase 2 (ERBB2), also known as HER2, is a transmembrane receptor that regulates cell proliferation, survival, and differentiation through activation of intracellular signaling pathways. Phospho-ErbB2 (pY1196) antibody, also referred to as phospho-HER2 antibody and phospho-ERBB2 antibody in the literature, detects phosphorylation at tyrosine 1196, a residue located within the intracellular domain of HER2 that is associated with sustained signaling activity.
Phosphorylation of HER2 at multiple intracellular residues creates a coordinated signaling network that governs receptor activation and downstream pathway engagement. Y1196 is positioned within this network and contributes to maintenance of signaling following initial receptor activation. While early phosphorylation sites initiate pathway engagement, residues such as Y1196 reflect continued signaling activity and support persistence of downstream responses.
HER2 signaling plays a central role in tumor biology, particularly in cancers characterized by ERBB2 amplification such as breast carcinoma. In these contexts, sustained signaling is critical for maintaining tumor cell proliferation and survival. Phosphorylation at Y1196 provides insight into ongoing receptor activity and supports evaluation of signaling persistence rather than initial activation alone.
Y1196 functions within a broader phosphorylation network that includes residues such as Y1112, Y1139, Y1221, Y1222, and Y1248. Together, these sites regulate progression of HER2 signaling from initiation through propagation to sustained activity. Analysis of phosphorylation at Y1196 complements detection of these other residues and contributes to a more complete understanding of HER2 signaling dynamics.
Phospho-specific detection of ERBB2 at Y1196 enables investigation of sustained receptor signaling and pathway maintenance. This supports studies focused on prolonged signaling, cellular adaptation, and response to therapeutic intervention.

This p-ERBB2 antibody was produced from rabbits immunized with a KLH conjugated synthetic phosphopeptide corresponding to amino acid residues surrounding pY1196 of human ERBB2.
Aliquot the p-ERBB2 antibody and store frozen at -20oC or colder. Avoid repeated freeze-thaw cycles.
