NUP62 Antibody Nuclear Pore Staining in HeLa Cells IF. Immunofluorescence staining of human HeLa cells with NUP62 Antibody / Nuclear Pore Complex Marker. Enzyme antigen retrieval was performed for 15 min, followed by blocking with 10% goat serum. The antibody was used at 1:50 overnight at 4oC, followed by a DyLight 488-conjugated goat anti-rabbit IgG secondary antibody at 1:500. Nuclei were counterstained with DAPI. NUP62 staining is observed at the nuclear periphery, consistent with its localization to nuclear pore complexes.

Description

NUP62 Antibody / Nuclear Pore Complex Marker detects nucleoporin 62 (NUP62), an FG-repeat-containing protein and core component of the nuclear pore complex (NPC). Nuclear pore complexes are large multiprotein assemblies embedded in the nuclear envelope that provide selective channels for the movement of proteins, RNA and other macromolecules between the nucleus and cytoplasm. NUP62 is associated with the central transport channel of the NPC and contributes to the selective permeability required for regulated nucleocytoplasmic transport.

NUP62 contains phenylalanine-glycine (FG) repeat sequences that interact with nuclear transport receptors as they move cargo through the nuclear pore. Together with other FG nucleoporins, NUP62 contributes to the permeability barrier that restricts passive movement of larger molecules while permitting receptor-mediated transport. NUP62 participates in a subcomplex with other nucleoporins, including NUP58 and NUP54, that helps form and organize the central channel of the nuclear pore complex.

Because of its integral association with the NPC, NUP62 is widely studied as a nuclear pore complex marker and as a component of the machinery controlling communication between nuclear and cytoplasmic compartments. NUP62 research is relevant to nuclear envelope organization, nuclear import and export, RNA trafficking, cell division and regulation of gene expression. Alterations involving nucleoporins and nuclear transport pathways are also investigated in cancer, neurological disease and other conditions associated with disrupted nuclear transport or nuclear pore function.

This recombinant rabbit monoclonal NUP62 Antibody has been validated by immunofluorescence in human HeLa cells and by Western blot across human, rat and mouse samples. Western blot testing includes human PC-3, HeLa, 293T and Jurkat cells, rat testis and RH35 cells, and mouse testis and NIH/3T3 cells. A prominent NUP62 band was observed at approximately 70 kDa across the tested samples. Immunofluorescence demonstrates NUP62 staining associated with the nuclear periphery in HeLa cells, consistent with its localization as a component of nuclear pore complexes.
