NLRP3 Antibody Cervical Carcinoma IHC. Immunohistochemistry analysis of FFPE human squamous cell cervical carcinoma tissue stained with NLRP3 Antibody. Cytoplasmic HRP-DAB brown staining is observed throughout tumor epithelial cell populations and adjacent inflammatory-associated cellular elements, consistent with the expected expression pattern of NLRP3 / Cryopyrin, an inflammasome-associated signaling protein involved in innate immune activation, inflammatory regulation, and pyroptosis-associated cellular pathways. Tissue sections underwent HIER using pH 8 EDTA buffer prior to immunostaining.
NLRP3 Antibody Tonsil IHC. Immunohistochemistry analysis of FFPE human tonsil tissue stained with NLRP3 Antibody. Scattered cytoplasmic HRP-DAB brown staining is observed within lymphoid and immune-associated cellular populations, consistent with the expected expression profile of NLRP3 / Cryopyrin, an inflammasome-associated signaling protein involved in innate immune activation, inflammatory regulation, and cytokine maturation-associated cellular pathways. Tissue sections underwent HIER using pH 8 EDTA buffer prior to immunostaining.
NLRP3 Antibody Multi-Species WB. Western blot analysis of 1) human ThP-1, 2) rat PC-12, 3) mouse spleen, and 4) mouse RAW264.7 cell lysates using NLRP3 Antibody. Bands are detected near approximately 110-120 kDa across multiple samples, consistent with the predicted molecular weight of NLRP3 / Cryopyrin, an inflammasome-associated signaling protein involved in innate immune activation, inflammatory regulation, cytokine maturation, and pyroptosis-associated cellular pathways.
NLRP3 Antibody ThP-1 FACS. Flow cytometry analysis of fixed and permeabilized human ThP-1 cells stained with NLRP3 Antibody (blue). Cells alone are shown in red and isotype control cells in green. The distinct fluorescence shift supports detection of endogenous NLRP3 / Cryopyrin expression in ThP-1 monocytic cells, consistent with the established role of this inflammasome-associated signaling protein in innate immune activation, inflammatory regulation, and cytokine maturation-associated cellular pathways.
NLRP3 Antibody Immune Cell Line WB. Western blot analysis of rat thymus tissue, mouse thymus tissue, mouse RAW264.7 cells, and mouse J774A.1 cells using NLRP3 antibody. Strong bands are detected near approximately 110-120 kDa across multiple immune-associated samples, consistent with the predicted molecular weight of NLRP3 / Cryopyrin, an inflammasome-associated signaling protein involved in innate immune activation, inflammatory regulation, cytokine maturation, and pyroptosis-associated cellular pathways. Additional lower molecular weight bands observed in selected macrophage-associated cell lines may reflect proteolytic processing or alternative inflammasome-associated protein species.
NLRP3 Antibody Colon Cancer IHC. Immunohistochemistry analysis of FFPE human colon cancer tissue stained with NLRP3 antibody. Predominantly cytoplasmic HRP-DAB brown staining is observed within stromal inflammatory-associated cellular populations adjacent to colonic epithelial structures, consistent with the established role of NLRP3 / Cryopyrin as an inflammasome-associated signaling protein involved in innate immune activation, inflammatory regulation, and cytokine maturation-associated cellular pathways. Tissue sections underwent HIER using pH 8 EDTA buffer prior to immunostaining.

Description

NOD-like receptor family pyrin domain containing 3 (NLRP3), also known as Cryopyrin or NALP3, is an intracellular pattern recognition receptor involved in inflammasome assembly, innate immune signaling, and inflammatory response regulation. NLRP3 Antibody / Inflammasome Marker is suitable for investigations involving inflammasome activation, innate immune signaling pathways, macrophage-associated inflammatory biology, and pyroptosis-associated cellular mechanisms. NLRP3 functions primarily within the cytoplasm, where it participates in multiprotein inflammasome complex formation in response to cellular stress and pathogen-associated danger signals.

NLRP3 antibody, also referred to as Cryopyrin antibody, NALP3 antibody, NOD-like receptor family pyrin domain containing 3 antibody, and Inflammasome signaling protein antibody in the literature, recognizes a central regulatory component of the NLRP3 inflammasome complex. Activation of NLRP3 promotes recruitment of ASC/PYCARD and caspase-1, resulting in maturation of inflammatory cytokines including IL1B and IL18. Through these pathways, NLRP3 contributes to inflammatory signaling, innate immune activation, and pyroptosis-associated cellular responses.

The NLRP3 inflammasome has attracted substantial interest in immunology and disease research because dysregulated inflammasome activation has been linked to autoinflammatory syndromes, neuroinflammatory disease, cardiovascular injury, metabolic inflammation, and infection-associated immune responses. NLRP3 signaling is widely studied in macrophages, monocytes, dendritic cells, and other innate immune-associated cellular populations. Because inflammasome activation influences multiple downstream inflammatory pathways, NLRP3 remains relevant for investigations involving cytokine maturation, inflammatory signaling regulation, and immune-associated disease biology.

Pattern recognition receptors such as NLRP3 participate in pathways controlling detection of pathogen-associated molecular patterns, sterile injury-associated danger signals, and intracellular stress-associated signaling events. NLRP3 activation has additionally been associated with oxidative stress responses, mitochondrial injury signaling, lysosomal disruption pathways, and ATP-responsive inflammatory signaling mechanisms. Through these interconnected regulatory systems, NLRP3 contributes to broader innate immune and inflammatory cellular environments.

Immunohistochemistry, western blot, and flow cytometry analyses support detection of endogenous NLRP3 expression in human cells and tissue, consistent with the expected cytoplasmic localization pattern of this inflammasome-associated signaling protein. The combined validation profile together with peer-reviewed publication support reinforces the utility of this rabbit polyclonal antibody for investigations involving inflammasome biology, innate immune signaling, pyroptosis-associated pathways, and inflammatory disease-associated cellular regulation.

An antibody targeting NLRP3 can therefore support studies involving inflammasome activation pathways, innate immune signaling regulation, cytokine maturation-associated biology, macrophage inflammatory responses, and pyroptosis-associated cellular mechanisms.

NLRP3 functions as a central regulator of inflammasome activation and innate immune signaling pathways; explore related inflammatory and immune regulatory proteins on our Immunology Antibodies page.

Application Notes

The stated application concentrations are suggested starting points. Titration of the NLRP3 antibody may be required due to differences in protocols and secondary/substrate sensitivity.
