MUC5AC Antibody 45M1 in Human Stomach IHC. Immunohistochemistry staining of FFPE human stomach tissue with MUC5AC Antibody 45M1 / Gastric and Respiratory Mucin Marker at 1-2 ug/ml for 30 min at RT. MUC5AC Antibody 45M1 / Gastric and Respiratory Mucin Marker detects MUC5AC, a large secreted gel-forming mucin produced predominantly by mucus-secreting epithelial cells of the stomach and respiratory tract. MUC5AC is a major structural component of mucus and contributes to the viscoelastic barrier that protects epithelial surfaces from pathogens, environmental particles, chemical injury and dehydration. Clone 45M1 is a widely published monoclonal antibody used to investigate MUC5AC expression in gastric, respiratory and tumor biology.

MUC5AC is extensively O-glycosylated and secreted into the extracellular environment, where mucin molecules assemble into hydrated mucus gels. In the stomach, MUC5AC is produced primarily by surface and foveolar epithelial cells and contributes to the protective mucus layer overlying the gastric mucosa. This barrier helps protect epithelial cells from gastric acid, digestive enzymes and microbial exposure. Changes in MUC5AC expression and glycosylation are studied in gastritis, gastric epithelial remodeling and interactions between the gastric mucosa and microorganisms such as Helicobacter pylori.

In the respiratory system, MUC5AC is produced by goblet and other mucus-secreting epithelial cells. Expression can increase in response to inflammatory and environmental stimuli, contributing to mucus hypersecretion and altered mucociliary clearance. MUC5AC is consequently investigated in airway inflammation, asthma, chronic bronchitis and chronic obstructive pulmonary disease. Regulation of MUC5AC production is also studied to understand epithelial responses to infection, allergens, cytokines and other stimuli that alter airway mucus production and composition.

MUC5AC expression is additionally relevant to cancer research. Altered mucin expression, localization and glycosylation occur in gastric adenocarcinoma and other epithelial malignancies, and MUC5AC can be investigated as part of the mucin phenotype of tumors. Clone 45M1 has been used extensively in studies involving gastric disease, oncology, respiratory biology and infectious disease. The V2198 datasheet demonstrates staining in normal human stomach, human gastric carcinoma and rat stomach, providing examples of MUC5AC detection in both normal and neoplastic gastric tissues.

Clone 45M1 is a mouse monoclonal antibody generated using an M1 mucin preparation from the fluid of an ovarian mucinous cyst as immunogen. It is reactive with human, mouse and rat samples and has been evaluated by immunohistochemistry in FFPE tissue. NSJ Bioreagents supplies antibodies for mucin, epithelial and cancer research. Application Notes

The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the MUC5AC Antibody 45M1 / Gastric and Respiratory Mucin Marker to be titered up or down for optimal performance.

1. Staining of formalin-fixed tissues requires boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 10-20 min followed by cooling at RT for 20 minutes.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

Immunogen

M1 mucin preparation from the fluid of an ovarian mucinous cyst belonging to an O Le(a-b) patient was used as the immunogen for this MUC5AC Antibody 45M1.

Storage

Store the MUC5AC Antibody 45M1 at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
