MUC5AC Antibody CLH2 in Human Gastric Carcinoma IHC. Immunohistochemistry staining of FFPE human gastric carcinoma tissue with MUC5AC Antibody CLH2 / Gastric Mucin Marker, clone CLH2. HIER was performed by steaming tissue sections in 10 mM Tris with 1 mM EDTA, pH 9, for 20 min followed by cooling before testing. MUC5AC staining is observed in gastric carcinoma cells, demonstrating detection of this gastric mucin marker in human tumor tissue.

MUC5AC Antibody CLH2 / Gastric Mucin Marker detects MUC5AC, a high molecular weight secreted glycoprotein and member of the gel-forming mucin family. MUC5AC is produced by specialized epithelial cells and is a major component of mucus covering the gastric mucosa. It is also expressed in tracheobronchial mucosa and other mucus-producing tissues. Through its extensive glycosylation and ability to form hydrated mucus gels, MUC5AC contributes to epithelial protection, lubrication and mucosal defense.

MUC5AC is prominently expressed by surface and foveolar epithelial cells of the stomach. Secreted MUC5AC contributes to the gastric mucus barrier that protects epithelial cells from acid, digestive enzymes, microorganisms and mechanical injury. Its characteristic distribution makes MUC5AC a useful gastric mucin marker for investigating mucus-producing epithelial cells, gastric differentiation and changes in mucosal architecture associated with disease.

Altered MUC5AC expression and glycosylation are studied in gastrointestinal disorders and cancer. Changes can accompany gastritis, intestinal metaplasia and neoplastic transformation of gastric epithelium. MUC5AC expression has also been investigated in pancreatic carcinoma and precursor lesions. In tumor pathology research, differential mucin expression can help characterize epithelial differentiation and mucinous tumor phenotypes. MUC5AC is frequently expressed in primary ovarian mucinous tumors but is generally absent from colorectal adenocarcinoma, where its expression pattern can complement markers such as MUC2 in studies of tumor origin.

Outside the gastrointestinal tract, MUC5AC is produced by mucus-secreting cells of the respiratory epithelium. Expression can increase in response to inflammation, infection and environmental stimuli, making the protein relevant to studies of airway mucus production and mucosal defense. These functions connect gastric and respiratory MUC5AC biology through their shared role in forming protective extracellular mucus barriers.

Clone CLH2 is a mouse monoclonal antibody generated against a synthetic peptide corresponding to the tandem repeat region of human MUC5AC. The antibody has been evaluated by immunohistochemistry in FFPE human gastric carcinoma tissue, supporting its use in studies of MUC5AC-positive epithelial cells and gastric tumor biology. 1. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

A synthetic peptide of human MUC5AC tandem repeat was used as the immunogen for the MUC5AC Antibody CLH2.

Storage

Store the MUC5AC Antibody CLH2 at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
