MUC5AC Antibody 9-13M1 / Gastric Mucin M1 Marker detects MUC5AC, a large secreted gel-forming mucin historically associated with gastric mucin M1. MUC5AC is produced by specialized mucus-secreting epithelial cells and is particularly abundant in the stomach and respiratory tract. Through extensive glycosylation and formation of hydrated mucus gels, MUC5AC contributes to epithelial protection, lubrication and mucosal defense against chemical, microbial and environmental challenges.
MUC5AC is prominently expressed by surface and foveolar epithelial cells of the stomach. Secreted MUC5AC contributes to the gastric mucus barrier that protects the underlying epithelium from acid, digestive enzymes and mechanical injury. Its characteristic distribution has made gastric mucin M1 and MUC5AC useful markers for investigating mucus-producing epithelial cells, gastric differentiation and changes in mucosal biology associated with disease.
Altered MUC5AC expression and glycosylation are investigated in gastrointestinal disorders and cancer. MUC5AC can be expressed in primary ovarian mucinous tumors while generally showing a different expression pattern from MUC2 in colorectal adenocarcinoma. Analysis of MUC5AC alongside other mucin and lineage markers can therefore contribute to studies of mucinous tumor differentiation. MUC5AC is also investigated in intestinal metaplasia, pancreatic carcinoma and precancerous changes involving pancreatic epithelium.
Outside the gastrointestinal tract, MUC5AC is produced by mucus-secreting cells of the respiratory epithelium. Its expression can increase in response to inflammatory signals, allergens, infection and environmental irritants. These functions connect gastric and respiratory MUC5AC biology through a common role in producing protective extracellular mucus barriers.
Clone 9-13M1 is a mouse monoclonal antibody that recognizes the peptide core of gastric mucin M1, now identified as MUC5AC. Its epitope is located within the N-terminal cysteine-rich region of the MUC5AC peptide core and is sensitive to beta-mercaptoethanol treatment. Clone 9-13M1 can also be paired with clone 1-13M1 for measurement of MUC5AC protein by ELISA. The antibody was generated using an M1 mucin preparation obtained from ovarian mucinous cyst fluid. An M1 mucin preparation from the fluid of an ovarian mucinous cyst belonging to an O Le(a-b) patient was used as the immunogen for the MUC5AC Antibody 9-13M1.
Store the MUC5AC Antibody 9-13M1 at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
