MR1 Antibody MR1/7579 Staining of Human Kidney IHC. Immunohistochemistry staining of FFPE human kidney tissue using MR1 Antibody clone MR1/7579. Heat-induced epitope retrieval was performed by boiling tissue sections in 10 mM Tris with 1 mM EDTA, pH 9, for 20 min followed by cooling. Strong brown immunoreactivity is evident in renal tubular epithelial cells, while glomeruli show comparatively limited staining. The inset shows the secondary antibody negative control with PBS substituted for the primary antibody.
Purified MR1 Antibody MR1/7579 SDS-PAGE. SDS-PAGE analysis of purified, BSA-free MR1 Antibody clone MR1/7579 under reduced (R) and non-reduced (NR) conditions with 2 ug loaded per lane. Under reducing conditions, bands corresponding to immunoglobulin heavy and light chains are visible at approximately 50 and 25 kDa, while the non-reduced antibody migrates predominantly near 150 kDa, consistent with intact IgG and supporting antibody integrity and purity.

Description

MR1 Antibody MR1/7579 / MR1 Antigen Presentation Antibody recognizes MR1, a non-classical major histocompatibility complex-related molecule specialized for the presentation of small-molecule antigens. MR1 associates with beta-2-microglobulin and presents metabolite-derived antigens to MR1-restricted T cells, particularly mucosal-associated invariant T (MAIT) cells. This function distinguishes MR1 from classical MHC class I molecules, which primarily present peptide antigens.

MR1 provides an important molecular connection between microbial metabolism and immune surveillance. Among the best-characterized MR1 ligands are intermediates derived from microbial riboflavin biosynthesis. When presented by MR1, these metabolites can be recognized by MAIT-cell T cell receptors and stimulate rapid immune responses. MR1-mediated antigen presentation therefore enables immune cells to detect metabolic signatures associated with a broad range of microorganisms.

MR1 trafficking and surface expression are closely regulated by ligand binding. In the absence of an appropriate ligand, much of the MR1 protein remains within intracellular compartments. Binding of suitable metabolites stabilizes MR1 and promotes trafficking of MR1-antigen complexes to the cell surface for recognition by MR1-restricted T cells. This regulated process makes an MR1 Antigen Presentation Antibody relevant to studies of metabolite sensing, antigen presentation and MAIT cell activation.

MR1-dependent immunity is studied extensively in host defense and mucosal immunology, while MR1-restricted T-cell responses are also being investigated in inflammation and cancer. Because MR1 is highly conserved and has limited polymorphism compared with classical MHC molecules, the MR1-MAIT cell axis represents a distinctive branch of cellular immunity with potential relevance across diverse disease settings.

Application Notes

Optimal dilution of the MR1 Antibody MR1/7579 / MR1 Antigen Presentation Antibody should be determined by the researcher.

Immunogen

A recombinant partial protein sequence (within amino acids 50-250) from the human protein was used as the immunogen for the MR1 antibody.

Storage

Aliquot the MR1 antibody and store frozen at -20oC or colder. Avoid repeated freeze-thaw cycles.

Alternate Names

MHC class I-related protein 1 antibody, Major histocompatibility complex class I-related protein 1 antibody, HLALS antibody
