MED28 Antibody in Human THP-1 Monocytic Leukemia Cells FACS. Flow cytometry analysis of fixed and permeabilized human THP-1 monocytic leukemia cells with MED28 Antibody / Mediator complex subunit 28 at 1 ug/million cells. Cells were blocked with 10% normal goat serum and incubated with the primary antibody for 30 min at 20oC. DyLight 488-conjugated goat anti-rabbit IgG was used as the secondary antibody at 5-10 ug/million cells for 30 min at 20oC. Red represents the unlabeled control, green represents the isotype control and blue represents MED28 Antibody staining. A clear fluorescence shift is observed with MED28 staining relative to the isotype control. This MED28 Antibody demonstrates detection of Mediator complex subunit 28 in human monocytic leukemia cells by flow cytometry.

MED28 Antibody in Human Leukemia and Hepatocellular Carcinoma Cells WB. Western blot testing of MED28 Antibody / Mediator complex subunit 28 using lysates from 1) human HEL cells, 2) human THP-1 cells, 3) human K562 cells and 4) human HepG2 cells. The antibody was used at 0.5 ug/ml overnight at 4oC, followed by goat anti-rabbit IgG-HRP at 1:5000 and chemiluminescent detection. A major band is detected at approximately 24 kDa, slightly above the predicted molecular weight of approximately 20 kDa and consistent with the reported migration of MED28. This MED28 Antibody demonstrates detection of Mediator complex subunit 28 in multiple human cell lines.

MED28 Antibody Immunoprecipitation from Human HepG2 Cells IP. Immunoprecipitation of MED28 from human HepG2 hepatocellular carcinoma whole cell lysate was performed using MED28 Antibody / Mediator complex subunit 28. Lane 1 contains 30 ug of HepG2 whole cell lysate, lane 2 is the rabbit IgG control and lane 3 contains MED28 immunoprecipitated from 500 ug of HepG2 lysate using 2 ug of antibody. Western blot detection was performed with MED28 Antibody at 0.5 ug/ml followed by goat anti-rabbit IgG-HRP and ECL detection. A major MED28 band is detected at approximately 24 kDa, slightly above its predicted molecular weight of approximately 20 kDa. This MED28 Antibody demonstrates immunoprecipitation of Mediator complex subunit 28 from human HepG2 cells.

MED28 Antibody in Human U2OS Osteosarcoma Cells IF. Immunofluorescence staining of human U2OS osteosarcoma cells with MED28 Antibody / Mediator complex subunit 28 at 5 ug/ml. Enzyme-mediated antigen retrieval was performed for 15 min, followed by blocking with 10% goat serum. Cells were incubated with the primary antibody overnight at 4oC. Fluoro488-conjugated goat anti-rabbit IgG was used as the secondary antibody at 1:500 for 30 min at 37oC. MED28 staining is predominantly localized to the nuclei. This MED28 Antibody enables visualization of Mediator complex subunit 28 expression and localization in human osteosarcoma cells.
