MBP Antibody / Brain Myelin Marker detects myelin basic protein (MBP), a major structural component of the myelin sheath surrounding neuronal axons. Encoded by the MBP gene, myelin basic protein contributes to the formation, compaction and stability of myelin membranes. In the central nervous system, MBP is produced primarily by oligodendrocytes and is widely used as a brain myelin marker for investigating myelination, glial cell development and nervous system organization.
MBP is a highly basic protein that associates with the cytoplasmic surfaces of myelin membranes. Its interactions with membrane lipids help bring adjacent layers together, producing the compact multilamellar structure required for normal myelin function. Alternative splicing of the MBP gene generates several protein isoforms that differ in molecular weight and developmental expression. These isoforms contribute to myelin assembly and maturation and can be detected as multiple bands when brain tissue is analyzed by Western blot.
As a brain myelin marker, MBP is commonly examined in the hippocampus, cerebral cortex, cerebellum and other regions of the central nervous system. Its expression provides a useful indicator of myelin distribution and can be studied alongside glial markers such as GFAP to investigate the relationship between myelinated structures and astrocytes. MBP is also relevant to studies of oligodendrocyte differentiation, developmental myelination and axon-glia interactions.
Changes in MBP expression, distribution or integrity are investigated in models of demyelination and remyelination, including research related to multiple sclerosis and experimental autoimmune encephalomyelitis. MBP is a recognized myelin-associated autoantigen and is also studied in neurological injury and neurodegenerative conditions involving changes to white matter or myelin integrity.
