Lambda Light Chain Antibody clone N10/2 is a mouse monoclonal reagent for detecting human immunoglobulins containing lambda light chains. Immunoglobulin molecules consist of two identical heavy chains and two identical light chains, with each antibody containing either kappa or lambda light chains. Lambda light chains contribute to the antigen-binding region of immunoglobulins and are expressed by a subset of mature B cells and antibody-secreting plasma cells.
Human lambda light chains are encoded by gene segments within the immunoglobulin lambda locus on chromosome 22. Rearrangement of variable and joining gene segments contributes to antibody diversity, while the resulting lambda chains associate with immunoglobulin heavy chains to form functional antibodies. Lambda light chains can occur within membrane-bound B-cell receptors and secreted immunoglobulins or as free immunoglobulin light chains.
Kappa and lambda light-chain expression is frequently examined when studying B-cell populations and immunoglobulin production. Individual B cells generally express immunoglobulins containing one light-chain class, whereas polyclonal populations contain both kappa- and lambda-expressing cells. A substantial restriction toward one light-chain class can therefore provide useful information when investigating clonal B-cell or plasma-cell populations.
Lambda light-chain detection is relevant to immunology, B-cell biology, antibody research, and studies of lymphoid and plasma-cell disorders. Free monoclonal immunoglobulin light chains have historically been referred to as Bence Jones proteins and are studied in connection with abnormal immunoglobulin production. Comparing lambda and kappa expression can further support research examining immunoglobulin light-chain distribution and B-cell clonality.
