JUNB Antibody PCRP-JUNB-3G2 in Human HeLa Cervical Cancer Cells IF. Immunofluorescence staining of PFA-fixed human HeLa cervical cancer cells with JUNB Antibody clone PCRP-JUNB-3G2 (green) and phalloidin counterstain (red). JUNB signal is visible primarily as fine punctate staining within the nuclei, with substantially less signal throughout the surrounding cytoplasm. Phalloidin delineates the actin cytoskeleton and cell boundaries, providing clear contrast with the predominantly nuclear JUNB distribution. This localization is consistent with the role of JunB as an AP-1 transcription factor and demonstrates cellular detection using this JunB Monoclonal Antibody.
JUNB Antibody PCRP-JUNB-3G2 in Human HeLa Cervical Cancer Cells FACS. Flow cytometry analysis of PFA-fixed human HeLa cervical cancer cells stained with JUNB Antibody clone PCRP-JUNB-3G2 (blue), compared with unstained cells (red). The antibody-stained population shows a clear rightward shift in fluorescence intensity relative to the negative control, demonstrating specific detection of endogenous JUNB in fixed HeLa cells. The well-separated fluorescence distributions support cellular detection of this AP-1 transcription factor using the JunB Monoclonal Antibody.
Purified JUNB Antibody PCRP-JUNB-3G2 SDS-PAGE. SDS-PAGE analysis of purified, BSA-free JUNB Antibody clone PCRP-JUNB-3G2 under reducing (R) and non-reducing (NR) conditions with a 2 ug antibody load. Under reducing conditions, the preparation resolves into the expected immunoglobulin heavy and light chains at approximately 50 kDa and 25 kDa, respectively. The non-reduced preparation shows a predominant intact antibody band at approximately 150 kDa. The expected electrophoretic profile provides a quality-control assessment of the integrity and purity of this JunB Monoclonal Antibody preparation.
JUNB Antibody PCRP-JUNB-3G2 Protein Microarray Validated. Protein microarray specificity testing of JUNB Antibody clone PCRP-JUNB-3G2 using a HuProt microarray containing more than 19,000 full-length human proteins. JUNB produced the highest signal with a Z-score of 132.08, compared with 20.81 for the next-ranked protein, resulting in an S-score of 111.26. The Z-score represents signal strength in standard deviations above the array mean, while the S-score represents the difference between the target and next-highest Z-scores. The exceptionally large separation of JUNB from all other proteins demonstrates strong target specificity of this JunB Monoclonal Antibody.

Description

JUNB Antibody PCRP-JUNB-3G2 / JunB Monoclonal Antibody recognizes JunB, a basic leucine zipper transcription factor encoded by the JUNB gene. JunB belongs to the JUN family of transcription factors together with c-Jun and JunD and participates in formation of activator protein-1 (AP-1) transcription factor complexes. AP-1 integrates signals from growth factors, cytokines, cellular stress, and other stimuli to regulate gene-expression programs involved in proliferation, differentiation, survival, inflammation, and tissue remodeling.

JunB contains a C-terminal basic region responsible for DNA binding and an adjacent leucine zipper that mediates dimerization. Through this domain, JunB can associate with other JUN proteins or members of the FOS family to form functional AP-1 complexes. These dimers recognize AP-1-responsive DNA sequences, including the TPA response element, and influence transcription according to the composition of the complex and cellular context. A JUNB Antibody can therefore be useful for investigating one specific component of the broader AP-1 regulatory network.

JUNB is an immediate-early response gene whose expression can change rapidly following extracellular stimulation. Unlike a simple constitutively expressed transcription factor, JunB participates in dynamic transcriptional responses that connect signaling pathways with changes in gene expression. Its activity is influenced by expression level, protein stability, post-translational modification, dimerization partners, and competition among AP-1 family members. These regulatory mechanisms allow JunB to produce context-dependent effects in different cell types and biological states.

JunB has important roles in cellular differentiation, proliferation, immune responses, bone biology, and tissue homeostasis. Altered JUNB regulation has also been investigated in cancer and other disorders involving abnormal growth or differentiation. Although JunB shares substantial sequence and functional relationships with other JUN family proteins, individual family members are not interchangeable. Their distinct regulatory regions and patterns of expression can produce different or even opposing transcriptional effects depending on biological context.

NSJ Bioreagents offers clone PCRP-JUNB-3G2 for research involving JunB and AP-1-associated transcriptional regulation. Protein microarray testing of this clone against a large collection of full-length human proteins provides an additional approach for assessing target selectivity. A JUNB Antibody such as clone PCRP-JUNB-3G2 provides a monoclonal reagent for investigating JunB expression and AP-1 biology, while a JunB Monoclonal Antibody provides an alternative protein-name terminology for studies of transcriptional regulation and cellular signaling.
