JAB1 (also known as COPS5) is a catalytic subunit of the COP9 signalosome (CSN), a conserved regulator of cullinâRING E3 ubiquitin ligases. Via its JAMM/MPN+ metalloprotease domain within the CSN, JAB1 promotes deneddylation of cullins, thereby tuning ubiquitin-dependent turnover of signaling and cell cycle regulators. JAB1 functions in both nucleus and cytoplasm, linking proteostasis to transcriptional responses and stress adaptation.
Through control of CRL activity, JAB1 influences pathways governing cell cycle progression, DNA damage responses, and transcription factor output (including AP-1).
