ITGB2 Antibody 1A3/2A10 recognizes integrin beta 2, a transmembrane adhesion receptor subunit encoded by the ITGB2 gene and commonly designated CD18. Integrin beta 2 is predominantly expressed by leukocytes and associates with several alpha integrin chains to form distinct beta 2 integrin heterodimers. These include LFA-1 (CD11a/CD18), Mac-1 (CD11b/CD18), complement receptor 4 (CD11c/CD18), and alpha D beta 2 (CD11d/CD18).
CD18 is a type I transmembrane glycoprotein with a large extracellular region, a single transmembrane segment, and a short cytoplasmic tail. Association with different alpha chains generates receptors with distinct ligand-binding properties. Conformational regulation of these heterodimers permits rapid changes in ligand affinity, while receptor engagement initiates intracellular signals and cytoskeletal rearrangements that regulate leukocyte adhesion and movement.
ITGB2-containing integrins interact with ligands involved in immune cell trafficking and host defense. LFA-1 recognizes intercellular adhesion molecules such as ICAM1, while other CD18-containing integrins participate in complement recognition, phagocytosis, and additional adhesive interactions. Through these heterodimers, CD18 contributes to firm leukocyte adhesion, transendothelial migration, immune synapse formation, inflammatory responses, and innate and adaptive immune functions.
The biological importance of ITGB2 is demonstrated by leukocyte adhesion deficiency type I, which results from pathogenic variants that impair CD18 expression or function. Loss of normal beta 2 integrin activity disrupts leukocyte adhesion and extravasation, limiting the ability of immune cells to migrate efficiently from the circulation into tissues. ITGB2 is consequently an important target in research involving leukocyte trafficking, inflammation, immune cell interactions, and adhesion disorders.
CD18 has historically been associated with LFA-1 nomenclature, but the ITGB2 protein is shared among multiple beta 2 integrins. Detection of CD18 therefore reflects expression of the common beta 2 subunit rather than specifically identifying the CD11a/CD18 LFA-1 heterodimer.
An ITGB2 Antibody can be used to investigate expression of the shared beta 2 integrin subunit, while a CD18 Monoclonal Antibody provides a monoclonal reagent for studies of leukocyte adhesion and integrin biology.
For a broader CD18-focused reagent, see our CD18 Antibody / ITGB2 Polyclonal Antibody page.
Optimal dilution of the ITGB2 Antibody 1A3/2A10 / CD18 Monoclonal Antibody should be determined by the researcher.
Human CD18 recombinant protein (amino acids Q404-S769) was used as the immunogen for the ITGB2 Antibody 1A3/2A10.
After reconstitution, the ITGB2 Antibody 1A3/2A10 can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.
