Isocitrate Dehydrogenase Antibody 16H7 / IDH1 Protein Antibody recognizes isocitrate dehydrogenase 1 (IDH1), an NADP-dependent enzyme found predominantly in the cytosol and also associated with peroxisomes. IDH1 catalyzes the oxidative decarboxylation of isocitrate to 2-oxoglutarate, also called alpha-ketoglutarate, while converting NADP+ to NADPH. This reaction connects IDH1 with cellular carbon metabolism, biosynthetic processes, and maintenance of cellular reducing capacity.
IDH1 is one of two NADP-dependent isocitrate dehydrogenases in mammalian cells. In contrast to mitochondrial IDH2, IDH1 functions primarily in the cytoplasmic compartment and contains a peroxisomal targeting sequence that supports localization to peroxisomes. Cytosolic Isocitrate Dehydrogenase contributes substantially to NADPH production, while peroxisomal IDH1 can provide reducing equivalents for metabolic reactions occurring within that organelle.
IDH1 is also extensively investigated in cancer biology because recurrent somatic mutations can alter its catalytic activity. Mutations involving arginine 132 are especially important and produce metabolic effects that differ from the normal function of IDH1. General detection of IDH1 protein therefore provides a complementary approach to mutation-specific reagents designed to recognize individual IDH1 variants.
