Intercellular Adhesion Molecule 1 Antibody 6F2C3 / LFA-1 Ligand Antibody recognizes ICAM1, a cell-surface glycoprotein and member of the immunoglobulin superfamily. Also known as ICAM-1 and CD54, ICAM1 contains five extracellular immunoglobulin-like domains, a single transmembrane region, and a short cytoplasmic domain. The protein functions as an adhesion molecule that helps coordinate interactions between leukocytes, vascular endothelial cells, and other cell types.
ICAM1 is a major ligand for lymphocyte function-associated antigen 1 (LFA-1), an integrin composed of CD11a and CD18. Binding between ICAM1 and LFA-1 promotes firm adhesion of leukocytes and contributes to their migration across vascular endothelium into surrounding tissues. ICAM1 can also bind Mac-1, composed of CD11b and CD18. Through these interactions, ICAM1 participates in leukocyte trafficking as well as stable contacts between lymphocytes and antigen-presenting cells during immune responses.
Expression of ICAM1 is regulated by inflammatory signals and can increase substantially following exposure to cytokines and other cellular stimuli. Elevated surface expression enhances the adhesive properties of endothelial, epithelial, and immune cells, facilitating leukocyte recruitment to sites of inflammation. ICAM1 has consequently been investigated in immune activation, inflammatory disorders, vascular biology, tissue injury, and cancer. Soluble forms of ICAM1 can also be released from cells and have been studied in relation to inflammatory and vascular processes.
ICAM1 additionally functions as the cellular receptor for major-group human rhinoviruses, connecting this adhesion molecule with host-pathogen interactions. Extensive N-linked glycosylation contributes to the substantially higher apparent molecular weight of mature ICAM1 compared with the predicted mass of its unmodified polypeptide.
