INI1 Antibody SMARCB1/3984 recognizes integrase interactor 1, a nuclear protein encoded by the SMARCB1 gene and also widely known as BAF47, SNF5 and hSNF5. SMARCB1 is a core component of the SWI/SNF family of ATP-dependent chromatin-remodeling complexes, which regulate access of transcriptional machinery and other nuclear factors to chromatin. Through these complexes, SMARCB1 participates in transcriptional regulation, cellular differentiation, proliferation and maintenance of normal developmental programs.
SMARCB1 functions as an important tumor suppressor. Biallelic inactivation of SMARCB1 is a defining molecular alteration in malignant rhabdoid tumors and is also associated with several other tumor types. Loss of functional SMARCB1 can disrupt normal SWI/SNF complex activity and alter transcriptional programs involved in cell-cycle regulation and differentiation. These properties have made INI1 expression an important subject of research in tumor biology and molecular pathology.
The protein is commonly referred to as INI1 because it was originally identified through its interaction with HIV-1 integrase. It is also known as BAF47, reflecting its association with the BAF form of the mammalian SWI/SNF complex, and as SNF5 or hSNF5 because of its relationship to the yeast SNF5 chromatin-remodeling protein. These names remain widely used in research and pathology literature, although SMARCB1 is the official gene symbol.
Altered or absent INI1 expression is particularly associated with malignant rhabdoid tumor, epithelioid sarcoma and several other uncommon neoplasms. Conversely, preserved nuclear expression can provide useful biological context when studying tumors that enter the differential diagnosis of SMARCB1-deficient neoplasms. Research into SMARCB1 loss has also contributed to a broader understanding of how disruption of SWI/SNF chromatin-remodeling complexes contributes to oncogenesis.
Because SWI/SNF complexes influence numerous transcriptional and developmental pathways, SMARCB1 is studied beyond its role as a tumor marker. Investigations of the protein encompass chromatin organization, epigenetic regulation, lineage specification, cell-cycle control and mechanisms of tumor suppression.
