IL-4 Antibody 11B11 recognizes interleukin 4, a secreted cytokine encoded by the IL4 gene that plays an important role in adaptive immunity and type 2 immune responses. IL-4 promotes the differentiation of naive CD4-positive T cells toward the T helper 2 phenotype and influences B-cell activation, proliferation, and antibody production. The cytokine was originally characterized through its effects on B cells and historically named B cell stimulatory factor 1 (BSF-1), reflecting an early understanding of its biological activity.
IL-4 signals through receptor complexes containing IL4RA (CD124). The type I IL-4 receptor combines IL4RA with the common gamma chain IL2RG, whereas the type II receptor pairs IL4RA with IL13RA1. Receptor engagement activates JAK-family kinases and downstream STAT6 signaling, producing transcriptional changes associated with type 2 immunity. IL-4 and IL-13 share the type II receptor complex, helping explain several overlapping biological effects of these cytokines.
IL-4 contributes to B-cell responses and immunoglobulin class switching, including mechanisms associated with IgE production. It also influences macrophage activation and communication among multiple immune-cell populations. These functions have made IL-4 an extensively studied cytokine in allergy, asthma, parasitic immunity, inflammation, autoimmunity, and other areas of immune regulation. Experimental manipulation of IL-4 activity has additionally helped researchers investigate how type 2 cytokine signaling affects immune responses under different physiological and disease conditions.
Clone 11B11 is a rat monoclonal antibody generated against partially purified native mouse IL-4 protein. This extensively published clone has been used in peer-reviewed research examining IL-4 biology and experimental modulation of IL-4 activity. As a B Cell Stimulatory Factor 1 Antibody, clone 11B11 provides a well-established monoclonal reagent for investigating the cytokine historically known as BSF-1.
