IL-2 Receptor Alpha Antibody in Mouse Spleen IHC. Immunohistochemistry of FFPE mouse spleen tissue using IL-2 Receptor Alpha Antibody demonstrates distinct membranous and cytoplasmic staining in a subset of cells distributed throughout the lymphoid tissue. HIER was performed by boiling tissue sections in pH 8 EDTA for 20 min followed by cooling before testing. The heterogeneous cellular staining demonstrates detection of CD25 by this CD25 Antibody in mouse spleen.
IL-2 Receptor Alpha Antibody in Rat Small Intestine IHC. Immunohistochemistry of FFPE rat small intestine tissue using IL-2 Receptor Alpha Antibody demonstrates prominent staining in cells within the intestinal mucosa, with a predominantly membranous and cytoplasmic pattern. IL-2 Receptor Alpha Antibody Detects CD25 in Mouse and Rat Samples WB. Western blot analysis using IL-2 Receptor Alpha Antibody detects a discrete band at approximately 45-50 kDa across mouse ANA-1, NIH 3T3, RAW264.7 and HEPA-1 cell lysates and rat C6, NRK and RH35 cell lysates (lanes 1-7, respectively). The observed migration is consistent with the reported 25-45 kDa range for variably glycosylated CD25, with modest sample-dependent differences in signal intensity. This multi-sample blot demonstrates detection of IL2RA by this CD25 Antibody in both mouse and rat samples.
IL-2 Receptor Alpha Antibody Detects CD25 in Mouse HEPA1-6 Hepatoma Cells FACS. Flow cytometry analysis of mouse HEPA1-6 hepatoma cells using IL-2 Receptor Alpha Antibody at 1 ug/million cells demonstrates a pronounced rightward fluorescence shift in the antibody-stained population (blue) relative to the isotype control (green) and unstained cells (red). Cells were blocked with goat serum before antibody staining. The clear population shift demonstrates cellular detection of CD25 by this CD25 Antibody in HEPA1-6 cells.

Description

IL-2 Receptor Alpha Antibody recognizes CD25, the alpha subunit of the interleukin 2 receptor encoded by the IL2RA gene. CD25 is a type I transmembrane glycoprotein involved in cellular responses to IL-2 and is expressed prominently by activated lymphocytes and regulatory T cells. Through its contribution to IL-2 receptor formation, CD25 participates in mechanisms controlling lymphocyte proliferation, survival, differentiation, and immune homeostasis.

CD25 can bind IL-2 independently as a low-affinity receptor but lacks the intracellular signaling capacity required to transmit the cytokine signal by itself. Association of CD25 with IL2RB (CD122) and the common gamma chain IL2RG (CD132) produces the high-affinity IL-2 receptor complex. Recruitment of IL-2 to this receptor facilitates signaling through pathways including JAK-STAT, PI3K-AKT, and MAPKs, allowing cells to respond efficiently to relatively low concentrations of IL-2.

IL2RA expression is induced following activation of conventional T and B lymphocytes and is constitutively high on many regulatory T cells. The relationship between CD25 and IL-2 is particularly important for regulatory T-cell development, maintenance, and suppressive function. CD25 is consequently studied in immune activation and tolerance, inflammatory responses, autoimmunity, and hematologic malignancies. Soluble CD25 can also be released from cells and has been investigated in the context of immune-system activation.

As a CD25 Antibody, it provides a reagent for investigating the alpha subunit of the IL-2 receptor and its relationship to lymphocyte activation and immune regulation.
