IDH1 Antibody IDH1/1152 / Cytosolic Isocitrate Dehydrogenase Antibody recognizes isocitrate dehydrogenase 1 (IDH1), a cytosolic and peroxisomal enzyme involved in cellular metabolism and redox regulation. IDH1 catalyzes the NADP-dependent oxidative decarboxylation of isocitrate to alpha-ketoglutarate, generating NADPH in the process. Through production of NADPH and alpha-ketoglutarate, IDH1 contributes to lipid biosynthesis, maintenance of cellular redox balance, and metabolic processes that support normal cell function.
IDH1 is distinct from the mitochondrial isocitrate dehydrogenases and functions primarily outside the mitochondrial tricarboxylic acid cycle. Cytosolic isocitrate dehydrogenase activity provides a source of NADPH used in reductive biosynthetic reactions and antioxidant defense. IDH1 therefore links intermediary metabolism with cellular responses to oxidative stress and changing metabolic demands. Alterations in IDH1 expression or activity can influence metabolic homeostasis, redox state, and the availability of metabolites used by other cellular pathways.
IDH1 is also an important target in cancer biology because recurrent somatic mutations occur at arginine 132. Mutant IDH1 proteins acquire neomorphic enzymatic activity that converts alpha-ketoglutarate to D-2-hydroxyglutarate, an oncometabolite capable of altering alpha-ketoglutarate-dependent cellular processes. IDH1 mutations are particularly associated with subsets of glioma, acute myeloid leukemia, chondrosarcoma, and other malignancies. Antibodies recognizing general IDH1 protein complement mutation-specific reagents by enabling investigation of IDH1 expression independently of a particular R132 substitution.

1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Tris with 1mM EDTA, pH 9.0 or 10mM Citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
