ICAM-3 Antibody 101-1D2 / ICAM3 D2 Domain Antibody recognizes ICAM3, a leukocyte cell-surface glycoprotein also known as CD50. ICAM3 belongs to the immunoglobulin superfamily of adhesion molecules and contains five extracellular immunoglobulin-like domains followed by a transmembrane region and cytoplasmic domain. Extensive N-linked glycosylation contributes to the mature protein migrating at approximately 120 kDa, substantially above the molecular weight predicted from its amino acid sequence.

ICAM3 is constitutively expressed at relatively high levels on resting leukocytes, including T lymphocytes, and contributes to adhesive interactions between immune cells. The protein is a major ligand for lymphocyte function-associated antigen 1 (LFA-1), the CD11a/CD18 integrin. The ICAM3-LFA-1 system participates in leukocyte adhesion and the establishment of cell-cell contacts associated with immune recognition and lymphocyte activation.

The extracellular region of ICAM3 contains five immunoglobulin-like domains that provide distinct structural and functional regions for molecular interactions. Clone 101-1D2 recognizes an epitope localized within the D2 extracellular domain of ICAM3. Published epitope-mapping experiments using ICAM3 domain-deletion constructs independently localized the 101-1D2 epitope to domain 2. In those experiments, 101-1D2 did not block binding of ICAM3-bearing cells to purified LFA-1, distinguishing its D2 epitope from several domain 1 epitopes involved more directly in LFA-1 recognition.

Clone 101-1D2 is a mouse monoclonal antibody originally generated against stimulated human leukocytes. The antibody has appeared in published ICAM3 research, including studies examining ICAM3 domain organization and adhesion-molecule interactions. For example, it has been used for ICAM3 detection on primary CD4-positive T cells and Jurkat T cells by flow cytometry. This research history provides useful clone-specific differentiation from other antibodies recognizing the same protein.

1. Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM Citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min.
