ICAM-1 Antibody W-CAM-1 / Wehi-CAM-1 Antibody recognizes intercellular adhesion molecule 1 (ICAM1), a cell-surface glycoprotein and member of the immunoglobulin superfamily. Also known as ICAM-1 and CD54, ICAM1 contains five extracellular immunoglobulin-like domains, a transmembrane region, and a cytoplasmic domain. It functions as an adhesion molecule that facilitates interactions among leukocytes, endothelial cells, and other cell types involved in immune surveillance and inflammatory responses.
ICAM1 is an important ligand for LFA-1, the CD11a/CD18 integrin expressed by leukocytes. ICAM1-LFA-1 interactions promote firm leukocyte adhesion and contribute to migration across vascular endothelium. ICAM1 also binds Mac-1, composed of CD11b and CD18, and participates in stable contacts between lymphocytes and antigen-presenting cells. These interactions place ICAM1 at an important junction between cell adhesion, leukocyte trafficking, and immune activation.
W-CAM-1 has a notable history in the ICAM1 literature. A 1988 study by Boyd and colleagues specifically identified the reagent as the Wehi-CAM-1 (W-CAM-1) monoclonal antibody and used it to investigate ICAM1 function. W-CAM-1 inhibited ICAM1-mediated cell aggregation, activated lymphocyte interactions, and T-cell adhesion to endothelial cells. The antibody also inhibited several immune processes dependent upon cell-cell contact, providing early functional evidence for the importance of ICAM1 in immune-cell adhesion. Subsequent work continued to identify the antibody as WEHI-CAM-1 or W-CAM-1 and described its generation following immunization with Raji Burkitt lymphoma cells.
ICAM1 is extensively N-glycosylated and mature protein can migrate at approximately 85-115 kDa, with variation related to cellular context and glycosylation. Clone 1H4 is also known as W-CAM-1 or Wehi-CAM-1, names that appear in the historical ICAM1 literature. Early studies using W-CAM-1 helped characterize the role of ICAM1 in cell adhesion and immune-cell interactions.

1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Tris with 1mM EDTA, pH 9.0 for 10-20 min followed by cooling at RT for 20 min
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

Store the ICAM-1 Antibody W-CAM-1 at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
