HSP72 Antibody / Inducible HSP70 Antibody recognizes the closely related stress-inducible proteins HSPA1A and HSPA1B. These members of the HSP70 molecular chaperone family are strongly induced by heat shock and other forms of cellular stress. HSP72 is commonly used to describe the inducible HSP70 proteins and distinguish them from constitutively expressed members of the broader HSP70 family.
HSPA1A and HSPA1B encode highly homologous proteins that help maintain proteome integrity during normal cellular activity and periods of stress. HSP70 chaperones bind exposed hydrophobic regions of unfolded or partially folded proteins, limiting inappropriate interactions and aggregation. An ATP-dependent cycle controls substrate binding and release, while numerous co-chaperones regulate HSP70 activity, substrate selection and interactions with other components of the protein quality-control machinery.
Inducible HSP70 expression can increase rapidly in response to elevated temperature, oxidative stress, metabolic disruption and other conditions that compromise protein stability. This response provides cells with additional chaperone capacity at times when damaged or unfolded proteins accumulate. HSPA1A and HSPA1B can assist protein refolding and cooperate with cellular degradation systems when damaged proteins cannot be restored, making HSP72 an important component of the proteotoxic stress response.
HSP72 biology is also closely connected with cell survival and apoptosis. Increased inducible HSP70 activity can help cells tolerate otherwise damaging conditions by stabilizing proteins and influencing stress-response pathways. These functions have generated substantial interest in HSP72 in cancer research, where tumor cells frequently encounter hypoxia, oxidative stress, nutrient limitation and elevated demands on protein quality-control systems.
NSJ Bioreagents offers this rabbit polyclonal HSP72 Antibody generated against a peptide corresponding to amino acids 577-596 of human HSPA1A. The immunogen sequence is also present in HSPA1B and therefore does not distinguish between these two highly related inducible HSP70 proteins. The stated application concentrations are suggested starting points. Titration of the HSP72 Antibody / Inducible HSP70 Antibody may be required due to differences in protocols and secondary/substrate sensitivity.
Amino acids 577-596 (VISWLDANTLAEKDEFEHKR-human) were used as the immunogen for this HSP72 antibody.
After reconstitution, the HSP72 antibody can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.
