HLA-G Antibody HLAG/6686 Staining of Human Tonsil Lymphoid Tissue IHC. Immunohistochemistry of FFPE human tonsil tissue using HLA-G Antibody clone HLAG/6686 at 2 ug/ml. Heat-induced epitope retrieval was performed by boiling tissue sections in 10 mM Tris containing 1 mM EDTA, pH 9, for 20 min followed by cooling before testing. Strong HLA-G immunoreactivity is observed across a broad population of cells within the lymphoid tissue.

HLA-G Antibody HLAG/6686 / MHC Class I Antigen G detects human leukocyte antigen G (HLA-G), a non-classical major histocompatibility complex class I molecule with important roles in immune regulation and tolerance. Unlike the broadly expressed classical HLA-A, HLA-B and HLA-C molecules, HLA-G has a relatively restricted physiological expression pattern. It was initially characterized in placental trophoblasts and is particularly associated with the maternal-fetal interface, where modulation of maternal immune responses contributes to tolerance of the developing fetus.

HLA-G shares the basic structural organization of MHC class I proteins and can associate with beta-2 microglobulin. Alternative splicing of HLA-G transcripts generates multiple membrane-bound and soluble isoforms, adding complexity to its immunoregulatory functions. Rather than functioning primarily as a conventional antigen-presenting molecule, HLA-G is recognized for its ability to influence both innate and adaptive immune responses. This distinctive biology has established MHC Class I Antigen G as an important target for studies of immune tolerance, pregnancy and immune-mediated disease.

HLA-G can regulate immune activity through interactions with inhibitory leukocyte immunoglobulin-like receptors, particularly LILRB1 (ILT2) and LILRB2 (ILT4). These receptors are distributed across multiple lymphoid and myeloid populations, allowing HLA-G to influence NK cells, T cells, B cells, monocytes, macrophages and antigen-presenting cells. HLA-G has also been studied in relation to KIR2DL4 on NK cells, although the biology of this interaction is complex. Through these receptor systems, HLA-G can contribute to suppression or modulation of cytotoxic and inflammatory immune responses.

The tolerogenic properties of HLA-G have generated considerable interest beyond reproductive immunology. Altered or induced HLA-G expression has been investigated in transplantation, inflammatory and infectious diseases, and cancer. In tumors, HLA-G expression is of particular interest as a potential mechanism of immune escape because suppression of immune effector activity may help malignant cells evade immune surveillance. HLA-G is therefore increasingly studied alongside immune checkpoint and other immunoregulatory molecules when examining interactions between tumor cells and the immune microenvironment.

Clone HLAG/6686 is a mouse monoclonal antibody directed against HLA-G. Its recognition of this non-classical MHC class I molecule provides a research reagent for investigating HLA-G protein expression and the biological contexts in which this immunoregulatory molecule is present. The HLA-G Antibody can complement antibodies to other HLA molecules and immune regulatory proteins when examining antigen presentation, immune recognition and mechanisms of immune tolerance.
