HLA-DRA Antibody / HLA-DR Alpha Chain Antibody recognizes HLA-DRA, the alpha-chain component of the HLA-DR major histocompatibility complex (MHC) class II molecule. HLA-DRA is encoded within the human MHC region on chromosome 6 and associates with an HLA-DR beta chain to form a membrane-anchored alpha/beta heterodimer. The extracellular domains of the assembled HLA-DR complex create a peptide-binding groove that accommodates peptides derived primarily from extracellular proteins. Presentation of these peptides to CD4-positive T cells provides a critical molecular connection between antigen recognition and adaptive immune responses.

HLA-DRA is the principal alpha chain used in HLA-DR complexes and can associate with beta chains encoded by HLA-DRB1 and, depending on the HLA haplotype, HLA-DRB3, HLA-DRB4 or HLA-DRB5. In contrast with the highly polymorphic HLA-DR beta chains, HLA-DRA exhibits comparatively limited polymorphism, particularly within regions contributing to the peptide-binding site. The HLA-DR alpha chain is a type I membrane glycoprotein with extracellular, transmembrane and cytoplasmic regions and has an approximate molecular weight of 33-35 kDa.

HLA-DR complexes are prominently expressed by professional antigen-presenting cells, including B lymphocytes, dendritic cells, monocytes and macrophages. Expression can also be induced in additional cell populations in response to inflammatory and immune signaling. Through its participation in HLA-DR complexes, HLA-DRA contributes to peptide presentation, CD4 T cell recognition and communication between innate and adaptive immune cells. HLA-DRA expression is consequently investigated in immunology, inflammation, infectious disease, autoimmunity, transplantation and cancer research.

Its targeting of the HLA-DR alpha-chain component distinguishes it from antibodies directed against individual polymorphic HLA-DR beta chains or conformational determinants of the assembled HLA-DR complex. An HLA-DR Alpha Chain Antibody can support research into MHC class II expression, antigen presentation and mechanisms through which HLA-DR contributes to CD4 T cell-mediated adaptive immunity.

Storage

After reconstitution, the HLA-DRA antibody can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.
