HLA-DR Antibody TAL 1B5 / HLA-DR Alpha Chain Antibody recognizes HLA-DR, a major histocompatibility complex (MHC) class II molecule central to antigen presentation and adaptive immunity. HLA-DR consists of an alpha chain encoded by HLA-DRA associated with a beta chain encoded by an HLA-DRB gene. Together, the two chains form a heterodimer that binds antigen-derived peptides for presentation to CD4-positive T cells, helping coordinate antigen-specific immune responses.

HLA-DR is prominently expressed by professional antigen-presenting cells, including B cells, monocytes, macrophages and dendritic cells, and expression can be induced in additional cell populations during immune activation. HLA-DR is consequently studied as both an antigen-presentation molecule and an immune-cell marker. Changes in HLA-DR expression are relevant to research involving inflammation, infectious disease, autoimmunity, transplantation and cancer, where MHC class II expression can influence interactions between antigen-presenting cells and helper T cells.

Clone TAL 1B5 is a well-established and extensively published monoclonal antibody with unusually detailed epitope characterization. Biochemical studies established that TAL 1B5 recognizes the HLA-DR alpha chain, and subsequent epitope-mapping experiments localized its binding site to the intracellular C-terminal tail of HLA-DRA. The antibody reacted with a synthetic peptide representing this region, while deletion of the C-terminal sequence eliminated TAL 1B5 recognition in Western blot and intracellular flow cytometry experiments. This defined molecular specificity provides particularly useful context for interpreting TAL 1B5 staining and distinguishes the clone from antibodies directed against extracellular or conformational HLA-DR determinants.

1. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
