Pan-HLA Class II Antibody HLA-Pan/8622R / HLA-DP/DQ/DR Antibody recognizes HLA-DP, HLA-DQ and HLA-DR, the three principal groups of classical human MHC class II molecules. These proteins enable antigen-presenting cells to display processed peptides to CD4-positive T lymphocytes, providing a critical step in the development of antigen-specific immune responses. MHC class II-mediated recognition influences helper T cell activation and subsequent regulation of B cells, macrophages and other components of immunity.
HLA class II molecules are composed of alpha and beta transmembrane glycoproteins that associate to form functional heterodimers. The extracellular regions of both chains contribute to the peptide-binding groove, while their membrane-spanning and cytoplasmic regions anchor the complex at the cell surface. Peptide loading occurs predominantly within specialized intracellular compartments following uptake and processing of extracellular antigens.
Professional antigen-presenting cells such as dendritic cells, B lymphocytes and macrophages constitutively express MHC class II molecules. Expression can also be induced in other cell populations during inflammatory responses. This regulated expression helps determine which cells can participate directly in class II-restricted antigen presentation and provides a mechanism through which cytokine signaling can modify interactions between immune cells and CD4-positive T lymphocytes.
Simultaneous recognition of HLA-DP, HLA-DQ and HLA-DR is useful when broad MHC class II detection is preferable to analysis of an individual HLA locus. Pan-HLA class II reagents can therefore complement locus-specific antibodies in studies comparing overall class II expression with the contributions of particular HLA molecules. MHC class II expression and regulation are relevant to research in immunology, inflammatory disease, transplantation, infection and tumor immunity.

Priess cells (a human B cell line) were used as the immunogen for the HLA-DP/DQ/DR antibody.
