HLA-ABC Antibody MHC-I/8147R / MHC Class I Antibody recognizes classical human major histocompatibility complex class I molecules represented by HLA-A, HLA-B and HLA-C. These highly polymorphic cell-surface glycoproteins are central components of adaptive immune surveillance. HLA class I molecules are expressed on nearly all nucleated cells and present intracellularly derived peptides to CD8-positive T lymphocytes, allowing the immune system to monitor cellular protein expression and identify cells affected by infection, malignant transformation or other intracellular abnormalities.
Classical MHC class I molecules consist of a polymorphic transmembrane heavy chain non-covalently associated with beta-2 microglobulin. The heavy chain contains alpha-1, alpha-2 and alpha-3 extracellular domains. The alpha-1 and alpha-2 domains form the peptide-binding groove, while beta-2 microglobulin contributes to structural stability of the assembled complex. HLA-A, HLA-B and HLA-C heavy chains are approximately 43-45 kDa glycoproteins, and their extensive genetic polymorphism enables recognition and presentation of a diverse repertoire of peptides.
Peptides presented by MHC class I molecules are generally generated through degradation of intracellular proteins. These peptides are transported into the endoplasmic reticulum by the transporter associated with antigen processing (TAP), where they are loaded onto newly assembled HLA class I molecules. Stable complexes containing the HLA heavy chain, beta-2 microglobulin and bound peptide subsequently pass through the Golgi apparatus to the plasma membrane. At the cell surface, peptide-HLA complexes can be surveyed by CD8-positive cytotoxic T cells.
HLA class I biology is important in research involving viral immunity, tumor immunology, transplantation, autoimmunity and immune evasion. Changes in HLA-A, HLA-B or HLA-C expression can influence antigen recognition by cytotoxic T cells, while class I molecules also participate in interactions with receptors expressed by natural killer cells. Consequently, altered MHC class I expression or antigen presentation can affect the balance between immune recognition and immune escape.
Clone MHC-I/8147R is a recombinant rabbit monoclonal antibody generated against a recombinant partial sequence within amino acids 100-300 of human HLA class I protein.
