Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. Histone H4 antibody, also referred to as H4 antibody or HIST1H4 antibody in the literature, is widely used to assess total histone content. The HIST1H4 gene family includes numerous variants that are co-expressed to meet the high demand for histone production during DNA replication. Detection across these variants ensures accurate measurement of histone H4 levels and avoids bias associated with gene-specific expression differences.

Structurally, all HIST1H4 gene products contribute to nucleosome formation by pairing with histone H3 to form the central tetramer that stabilizes DNA wrapping.
