HGF Antibody in Human, Rat and Mouse Liver and Cell Lysates WB. Western blot testing of HGF Antibody / MET Receptor Ligand Antibody in 1) human hepatocellular carcinoma tumor tissue, 2) human hepatocellular carcinoma paracancerous tissue, 3) human HepG2 cells, 4) human HUH7 cells, 5) rat liver, 6) rat RH35 cells and 7) mouse liver. The HGF single-chain precursor has a predicted molecular weight of ~83 kDa. A predominant band is observed at ~70 kDa, with additional species at ~50-60 kDa in some samples, consistent with the known proteolytic processing and glycosylation-dependent migration of HGF.

Description

HGF Antibody / MET Receptor Ligand Antibody recognizes hepatocyte growth factor, a multifunctional secreted protein encoded by the HGF gene. HGF was originally characterized through its ability to stimulate hepatocyte proliferation and was subsequently shown to be identical to scatter factor, a protein identified independently through its effects on epithelial cell motility. HGF functions as a mitogen, motogen and morphogen and has important roles in tissue development, regeneration and cellular responses to injury.

HGF is synthesized as an inactive single-chain precursor and undergoes proteolytic processing to generate the biologically active two-chain form. Structurally, the protein contains an N-terminal domain, four kringle domains and a C-terminal serine protease homology domain, although HGF itself lacks protease activity. This multidomain organization enables interactions required for receptor binding and activation.

The receptor for HGF is the MET receptor tyrosine kinase. Binding of HGF to MET initiates signaling pathways that regulate proliferation, survival, migration and morphogenesis. HGF-MET signaling is particularly important in epithelial-mesenchymal interactions and contributes to processes such as embryonic development and tissue regeneration. Dysregulation of the HGF-MET axis has also been extensively investigated in cancer, where aberrant pathway activity can contribute to tumor growth, invasive behavior and metastatic progression.

This rabbit polyclonal antibody was generated against a recombinant fragment of human HGF and is antigen affinity purified. NSJ Bioreagents provides this HGF Antibody for research into growth factor expression, MET signaling, tissue regeneration and cancer biology. A MET Receptor Ligand Antibody can support studies examining the expression and biological regulation of HGF within this important growth factor signaling pathway.

Application Notes

The stated application concentrations are suggested starting amounts. Titration of the HGF Antibody / MET Receptor Ligand Antibody may be required due to differences in protocols and secondary/substrate sensitivity.

Immunogen

An amino acid sequence from the N-terminus of mouse HGF (QKKRRNTLHEFKKSAKTTLTKEDPLLKIKTKK) was used as the immunogen for this HGF antibody.

Storage

After reconstitution, the HGF antibody can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.

Alternate Names

HGF antibody, hepatocyte growth factor antibody, scatter factor antibody, SF antibody, hepatopoietin A antibody
