Gelsolin Antibody / Actin Severing Protein Antibody recognizes GSN, a calcium-regulated actin-binding protein that controls the assembly and disassembly of actin filaments. Gelsolin is a prominent member of the gelsolin/villin superfamily and functions through actin filament severing, capping and nucleation. By remodeling filamentous actin, GSN contributes to dynamic changes in cell shape, motility, adhesion, migration and other processes dependent on cytoskeletal organization.
Gelsolin exists in intracellular and secreted forms generated through alternative transcription initiation and splicing. Cytoplasmic gelsolin participates directly in actin cytoskeleton remodeling, while plasma gelsolin contributes to the extracellular actin-scavenging system. Following tissue injury or cell death, extracellular actin can accumulate in the circulation, where plasma gelsolin binds and clears actin filaments. These distinct forms make gelsolin relevant to research involving both intracellular cytoskeletal dynamics and extracellular responses to tissue damage.
Beyond its structural role, GSN has been investigated in cell survival, apoptosis and cancer biology. Interactions between gelsolin-regulated actin remodeling and signaling pathways can influence cell migration, invasion and resistance to apoptotic stimuli. Altered gelsolin expression has been reported in multiple tumor types, making the protein useful for studies of cancer cell behavior and cytoskeletal reorganization. Pathogenic variants in GSN are also associated with gelsolin amyloidosis, historically known as familial amyloidosis of the Finnish type.
This rabbit polyclonal antibody was generated against a peptide corresponding to amino acids 763-775 of human gelsolin and is antigen affinity purified. The immunizing sequence is conserved in rat gelsolin, and the antibody is reactive with human, mouse and rat samples. Amino acids 763-775 (WDDDYWSVDPLDR-human) were used as the immunogen for this Gelsolin antibody (100% rat homology).
After reconstitution, the Gelsolin antibody can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.
