Friend leukemia integration 1 (FLI1) is a nuclear ETS family transcription factor involved in hematopoietic differentiation, endothelial development, and transcriptional regulation of lineage-associated gene expression pathways. FLI1 expression is commonly associated with endothelial cells, hematopoietic progenitors, megakaryocytes, and selected lymphoid populations. Through transcriptional regulation of lineage-associated target genes, FLI1 contributes to cellular differentiation and maintenance of specialized developmental programs.
ETS family transcription factors participate in pathways controlling proliferation, differentiation, migration, angiogenesis, and developmental signaling. FLI1 has additionally attracted substantial interest in oncology because chromosomal translocations involving FLI1 are characteristic of Ewing sarcoma family tumors. Fusion proteins involving EWSR1 and FLI1 generate aberrant transcriptional programs that contribute to tumor development and oncogenic signaling pathways.
