EIF2S3 Antibody / Eukaryotic translation initiation factor 2 subunit 3 Antibody recognizes the gamma subunit of eukaryotic translation initiation factor 2, a central component of the machinery that begins protein synthesis. The EIF2S3 gene encodes the GTP-binding subunit of the eIF2 heterotrimer, which also contains alpha and beta subunits. During initiation, eIF2 binds GTP and initiator methionyl-tRNA to form a ternary complex. This complex delivers the initiator tRNA to the small ribosomal subunit as the ribosome prepares to identify a translation start codon.
The gamma subunit supplies the nucleotide-binding core and contributes to the recognition of initiator tRNA. Once the scanning ribosomal complex identifies an appropriate start site, GTP hydrolysis and subsequent factor release permit progression toward assembly of an elongation-competent ribosome. The nucleotide state of eIF2 must then be reset by the guanine nucleotide exchange factor eIF2B. These linked reactions make initiation sensitive to both the availability of charged initiator tRNA and the regulatory state of the eIF2 system.
The EIF2S3 Antibody recognizes the gamma subunit, which performs the nucleotide-binding function that distinguishes its contribution from those of the alpha and beta subunits. One major control mechanism is the integrated stress response. Cellular stresses activate kinases that phosphorylate the alpha subunit of eIF2, converting the complex into an inhibitor of productive nucleotide exchange by eIF2B. This reduces overall initiation while favoring translation of selected transcripts with specialized regulatory features, including ATF4. Although EIF2S3 itself is not the principal phosphorylation target, the activity of its GTP-binding subunit is inseparable from this regulated cycle. Changes in ternary-complex availability can therefore reshape protein synthesis across the cell.
Pathogenic EIF2S3 variants cause an X-linked neurodevelopmental disorder commonly associated with MEHMO syndrome, which can involve microcephaly, epilepsy, developmental impairment, hypogonadism, and obesity. These phenotypes show that even a broadly required translation factor can have especially pronounced consequences in developing tissues. The relationship between particular EIF2S3 variants, initiation efficiency, and tissue-specific vulnerability remains important for understanding how altered protein synthesis produces complex developmental disease.
