E-cadherin Antibody Colon Carcinoma IHC. Immunohistochemistry analysis of Cadherin 1 / CDH1 expression in FFPE human colon carcinoma using HuProt specificity validated clone CDH1/1525 antibody, showing membranous HRP-DAB brown staining in tumor epithelial cells with preserved cell-cell junction localization, while surrounding stromal components remain largely negative. Signal highlights epithelial differentiation and junctional organization within carcinoma tissue.
Formalin-fixed, paraffin-embedded human colon carcinoma stained with E-Cadherin antibody (CDH1/1525).
Formalin-fixed, paraffin-embedded human prostate carcinoma stained with E-Cadherin antibody (CDH1/1525).
Formalin-fixed, paraffin-embedded human skin stained with E-Cadherin antibody (CDH1/1525).
Formalin-fixed, paraffin-embedded human placenta stained with E-Cadherin antibody (CDH1/1525).
Western blot analysis A) partial recombinant protein B) human stomach lysate using E-Cadherin antibody (CDH1/1525). Expected molecular weight: 135 kDa (precursor), 80-120 kDa (mature, depending on glycosylation level).
Immunofluorescent staining of human MCF7 cells with HuProt specificity validated E-Cadherin antibody (clone CDH1/1525, green) and Reddot nuclear stain (red).
E-cadherin Antibody HuProt Specificity Validation. Proteome-wide microarray analysis of Cadherin 1 / CDH1 specificity using clone CDH1/1525 antibody on a HuProt(TM) array containing over 19,000 full-length human proteins, demonstrating strong and selective binding to CDH1 as the top-ranked target with a dominant Z-score and high S-score relative to non-target proteins. The Z-score represents signal intensity in standard deviations above the array mean, while the S-score reflects the difference between the top-ranked protein and subsequent hits, indicating high relative specificity. These results support highly selective recognition of CDH1 with minimal off-target binding across the human proteome.
SDS-PAGE analysis of purified, BSA-free E-Cadherin antibody (clone CDH1/1525) as confirmation of integrity and purity.
E-cadherin Breast Adenocarcinoma IHC. Immunohistochemistry of formalin-fixed, paraffin-embedded human breast adenocarcinoma stained with E-Cadherin antibody (CDH1/1525).

Description

E-cadherin (CDH1) is a transmembrane adhesion protein that plays a central role in epithelial cell-cell interactions and maintenance of tissue architecture. E-cadherin Antibody / HuProt Specificity Validated Antibody (clone CDH1/1525) detects this protein with specificity supported by HuProt microarray screening, a large-scale platform containing thousands of full-length human proteins. E-cadherin is also widely known as Cadherin 1 antibody and is a key marker of epithelial differentiation and structural integrity.

HuProt microarray validation represents a comprehensive approach to antibody specificity assessment by evaluating binding across a near-complete human proteome in parallel. This platform enables identification of target recognition relative to a wide array of structurally diverse proteins, providing a high level of confidence in specificity. Clone CDH1/1525 antibody is therefore positioned as a rigorously evaluated reagent for studies requiring extensive validation against potential off-target interactions.

In research applications, accurate detection of CDH1 is essential for investigating epithelial biology, tumor progression, and cellular differentiation. Changes in E-cadherin expression or localization are closely associated with epithelial-to-mesenchymal transition and altered adhesion states. A HuProt specificity validated E-cadherin antibody supports reliable interpretation of these changes by minimizing the likelihood of non-specific signal in complex biological samples.

E-cadherin functions through interactions with catenins and cytoskeletal components to regulate adhesion, polarity, and intracellular signaling pathways. These interactions are critical for maintaining epithelial structure and coordinating cellular behavior. Reliable detection of CDH1 supported by HuProt validation enhances the ability to study these processes across diverse experimental systems, including cultured cells and tissue-derived samples.

The mouse monoclonal clone CDH1/1525 antibody supports consistent recognition of CDH1 and is particularly well suited for applications requiring high specificity validation against a large panel of human proteins. This E-cadherin antibody is especially valuable for studies where rigorous specificity assessment is important, including experiments involving complex proteomes or closely related protein families.

Compared to standard protein microarray approaches, HuProt validation provides deeper coverage of the human proteome and enables more comprehensive evaluation of antibody binding behavior. This distinction supports positioning of clone CDH1/1525 as a high-confidence reagent for epithelial marker detection in demanding experimental contexts.

This antibody is part of the CDH1 antibody collection, where multiple E-cadherin antibody formats and applications are available for studying epithelial adhesion and cancer progression.

Application Notes

Optimal dilution of the E-cadherin Antibody / HuProt Specificity Validated Antibody should be determined by the researcher.

1. Staining of formalin-fixed tissues requires boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 10-20 min followed by cooling at RT for 20 min.

Immunogen

A full length recombinant human protein was used as the immunogen for the E-Cadherin antibody.

Storage

Store the E-Cadherin antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).

Alternate Names

CDH1 antibody, E-cadherin antibody, Cadherin 1 antibody, epithelial cadherin antibody, adherens junction protein antibody
