DNAJB9 Antibody / DnaJ homolog subfamily B member 9 Antibody targets an endoplasmic reticulum (ER) co-chaperone involved in protein folding and the response to disturbances in ER homeostasis. DNAJB9, also called ERdj4, belongs to the Hsp40/DnaJ family and contains a J domain that helps regulate the ATPase cycle of its Hsp70 partner, the ER chaperone BiP. By cooperating with BiP, DNAJB9 participates in the handling of proteins that have not achieved their correct conformations within the secretory pathway.
The ER must fold proteins destined for secretion, membranes, and several intracellular compartments. Misfolded proteins expose regions that are normally buried, creating a risk of inappropriate aggregation. ER-associated chaperones bind such intermediates and promote productive folding or direct persistently defective proteins toward quality-control pathways. DNAJB9 is associated with ER-associated degradation, a process in which selected misfolded substrates are ultimately transported out of the ER and destroyed by the proteasome. Its interaction with BiP links substrate recognition to chaperone-mediated quality control.
DNAJB9 expression increases during the unfolded protein response, particularly through transcriptional programs activated when misfolded proteins accumulate. This induction helps expand the ER's capacity to manage proteotoxic stress. However, sustained ER stress can also affect translation, metabolism, and cell survival, so the consequences of DNAJB9 regulation depend on the duration and severity of the underlying disturbance.
DNAJB9 has additional significance in fibrillary glomerulonephritis, a kidney disorder characterized by distinctive fibrillar deposits. Strong DNAJB9 accumulation is a characteristic molecular finding in this disease, although the mechanisms connecting the protein to deposit formation remain under investigation. Its prominence in this setting has renewed interest in functions beyond conventional ER quality control.
NSJ Bioreagents offers this DNAJB9 Antibody to help examine how ER chaperone networks respond to misfolded proteins and how DNAJB9 biology intersects with abnormal protein deposition. The protein provides a useful point of connection between intracellular proteostasis, adaptive stress signaling, and disease-associated changes in protein organization.
