DNAJB12 Antibody by FCM in Fixed A431 Cells. Flow cytometry analysis of fixed and permeabilized A431 cells stained with DNAJB12 Antibody shows a rightward shift of the blue antibody-stained histogram relative to the green isotype control. The red histogram represents the unstained blank. Cells were fixed with 4% paraformaldehyde and permeabilized before intracellular labeling; this experiment does not represent live-cell surface staining.
DNAJB12 Antibody by FCM in Fixed U251 Cells. Flow cytometry analysis of fixed and permeabilized U251 cells stained with DNAJB12 Antibody. The blue test histogram shifts to higher fluorescence than the green isotype control, with the red blank histogram at lower fluorescence. Intracellular staining was performed after fixation and permeabilization.
DNAJB12 Antibody by IHC in Human Endometrial Cancer Tissue. Immunohistochemistry of a paraffin-embedded human endometrial cancer section with DNAJB12 Antibody shows brown DAB staining in malignant glandular epithelial cells, with a predominantly cytoplasmic distribution. Hematoxylin counterstaining highlights blue nuclei; connective tissue is less intensely labeled.
DNAJB12 Antibody by IHC in Human Stomach Cancer Tissue. Immunohistochemistry of a paraffin-embedded human stomach cancer section using DNAJB12 Antibody reveals heterogeneous brown DAB labeling among gland-forming tumor cells. Hematoxylin-stained nuclei remain blue-purple, while cytoplasmic staining varies across the field.
DNAJB12 Antibody by IHC in Human Stomach Tissue. Immunohistochemistry of a paraffin-embedded human stomach section using DNAJB12 Antibody shows brown DAB immunoreactivity in glandular epithelial cells, including punctate and granular areas of staining. Blue hematoxylin counterstaining distinguishes nuclei from the surrounding cytoplasm and connective tissue.
DNAJB12 Antibody by IF in U2OS Cells. Immunofluorescence microscopy of U2OS cells labeled with DNAJB12 Antibody shows green fluorescence concentrated in the cytoplasm and around the nuclear periphery. Cell nuclei are counterstained blue. The images do not establish localization to a specific organelle without a colocalization marker.
DNAJB12 Antibody by WB in Human and Rodent Lysates. Western blot analysis using DNAJB12 Antibody shows immunoreactive bands near 36-40 kDa in eight samples. Lanes 1-4 contain HaCaT, U251, A431, and Caco-2 human whole-cell lysates; lanes 5-6 contain rat kidney and lung tissue; lanes 7-8 contain mouse kidney and lung tissue. The predominant band is close to the expected molecular weight for DNAJB12, although its intensity varies among samples. Additional faint lower bands are visible in some lanes but cannot be assigned a molecular identity from this blot alone. The result supports detection in the tested human and rodent preparations.

Description

DNAJB12 Antibody / DnaJ homolog subfamily B member 12 Antibody targets DNAJB12, an endoplasmic reticulum membrane-associated member of the Hsp40/DnaJ cochaperone family. DNAJB12 contains a J domain that stimulates the ATPase cycle of Hsp70 chaperones, linking it to protein quality-control pathways. The endoplasmic reticulum is responsible for folding and processing many membrane and secreted proteins, and proteins that fail to achieve an appropriate conformation must be retained, refolded, or removed. DNAJB12 participates in this surveillance environment.

Unlike soluble cochaperones, DNAJB12 is anchored to the ER membrane, placing its chaperone-regulatory activity near substrates and machinery involved in membrane protein biogenesis. J-domain proteins generally determine where and when Hsp70 systems engage client proteins, while Hsp70 uses ATP-dependent cycles to bind and release exposed polypeptide regions. DNAJB12 has been linked to ER-associated degradation, the pathway that directs selected misfolded proteins toward extraction from the ER and destruction by the proteasome. Its activity must be interpreted alongside other chaperones and ubiquitin-associated factors that collectively determine substrate fate.

ER protein quality control balances two competing needs: preventing the accumulation of potentially harmful misfolded species and preserving proteins that can still reach their functional conformations. Altering the activity of a cochaperone may shift this balance without necessarily changing the overall capacity for protein synthesis. DNAJB12 is therefore relevant to understanding how membrane-associated chaperone networks discriminate among folding intermediates. NSJ Bioreagents offers this DNAJB12 Antibody to support investigation of ER chaperone organization and the handling of proteins that encounter folding difficulties during biosynthesis. Identifying the clients and partner proteins most dependent on DNAJB12 may reveal how localized Hsp70 regulation contributes to proteostasis.
