DNAJB11 Antibody / DnaJ homolog subfamily B member 11 Antibody detects DNAJB11, an endoplasmic reticulum-associated Hsp40 co-chaperone, also known as ERdj3. It belongs to the DnaJ family of proteins that cooperate with Hsp70 chaperones to assist protein folding. DNAJB11 operates within the secretory protein quality-control system, where newly synthesized polypeptides must attain appropriate conformations before leaving the endoplasmic reticulum. Its J domain stimulates the ATPase activity of the ER Hsp70 chaperone BiP, also called HSPA5 or GRP78. This interaction helps regulate the binding and release of client proteins during their maturation.
The endoplasmic reticulum contains specialized machinery for folding secreted and membrane-associated proteins, forming disulfide bonds, and recognizing improperly folded molecules. DNAJB11 participates in this environment by engaging unfolded substrates and supporting their handling by BiP. The protein has also been described as a soluble ER luminal co-chaperone, rather than a component of the mitochondrial DnaJ system. Its activities intersect with the unfolded protein response, a network that adjusts ER protein-folding capacity when the burden of misfolded proteins increases. The outcome of these responses depends on the severity and duration of ER stress. The DnaJ homolog subfamily B member 11 Antibody designation refers to this same protein target.
Pathogenic variants in DNAJB11 have been associated with a form of autosomal dominant tubulointerstitial kidney disease featuring cystic and fibrotic changes. This clinical association underscores the importance of ER proteostasis for long-lived epithelial cells. DNAJB11-related disease differs from classical polycystic kidney disease caused by defects in primary ciliary signaling, even when some clinical features overlap. Research on DNAJB11 therefore connects molecular chaperone biology with the maintenance of organelle function and tissue integrity. NSJ Bioreagents includes DNAJB11 among proteins involved in ER folding and quality control. A DNAJB11 Antibody provides a means of examining the molecular functions associated with DNAJB11.
