DHX16 Antibody / DEAH-box helicase 16 Antibody targets an ATP-dependent RNA helicase involved in pre-messenger RNA splicing. DHX16 is a member of the DEAH-box family of proteins, whose conserved helicase cores couple nucleotide hydrolysis to changes in RNA-protein interactions. Splicing removes introns from newly transcribed RNA and joins exons to produce mature transcripts. This process requires a spliceosome that repeatedly rearranges its RNA and protein components as it progresses through assembly, catalysis, and disassembly.
DHX16 is closely related to the yeast splicing factor Prp2, an ATPase involved in remodeling the spliceosome before the first catalytic step. Such remodeling is essential because the spliceosome initially assembles in a configuration that is not yet competent to catalyze intron removal. ATP-driven rearrangements help expose or reposition RNA elements and displace proteins that would otherwise obstruct catalysis. DHX16 is therefore associated with a crucial transition between spliceosome assembly and productive RNA processing.
DEAH-box helicases are sometimes described as RNA unwinding enzymes, but their cellular activities often involve remodeling ribonucleoprotein complexes rather than simply separating complementary RNA strands. Their function depends on interactions with other splicing factors and the structural state of the spliceosome. For DHX16, identifying these interactions is important for distinguishing direct remodeling events from secondary consequences of stalled splicing.
Accurate intron removal is necessary to maintain coding sequences and regulate transcript diversity. Failure at a catalytic transition can cause retention of introns, abnormal splice products, or degradation of incompletely processed RNA. Because many genes rely on coordinated splicing, disruption of a central remodeling factor can affect numerous downstream cellular pathways.
NSJ Bioreagents offers this DHX16 Antibody to facilitate investigation of ATP-dependent spliceosome remodeling and the events that permit pre-mRNA splicing to proceed. Understanding DHX16's position in the splicing cycle can help clarify how dynamic RNA-protein assemblies achieve the precision needed for reliable gene expression.
