Desmin Antibody D33 is a mouse monoclonal antibody recognizing desmin, a type III intermediate filament protein that forms an essential component of the muscle cell cytoskeleton. Desmin is expressed predominantly in skeletal, cardiac, and smooth muscle cells, where it contributes to structural integrity and the organization of contractile elements. Its characteristic distribution makes desmin an important marker of muscle differentiation in normal tissues and muscle-derived tumors. The D33 monoclonal antibody is a longstanding reagent in muscle biology and immunohistochemical research, particularly for identifying cells exhibiting myogenic differentiation.
The human DES gene is located on chromosome 2q35 and encodes a protein of approximately 53 kDa comprising 470 amino acids. Desmin contains a central alpha-helical rod domain flanked by nonhelical head and tail regions. These structural features support the assembly of desmin into intermediate filaments that form an interconnected cytoskeletal network. In striated muscle, desmin filaments surround the Z discs of sarcomeres and help maintain the alignment of adjacent myofibrils. The filaments also provide structural connections between the contractile apparatus, mitochondria, nuclei, and plasma membrane.
Desmin expression is closely associated with muscle cell differentiation and maturation. During myogenesis, desmin becomes an important component of the developing cytoskeleton as precursor cells acquire specialized contractile functions. Its expression persists in differentiated skeletal, cardiac, and smooth muscle cells. The Muscle-Specific Intermediate Filament Antibody is therefore relevant to investigations of muscle development, cytoskeletal organization, tissue remodeling, and regeneration. Desmin is also frequently studied alongside other muscle-associated markers to characterize cellular differentiation and structural changes in muscle tissue.
Desmin is particularly valuable in research involving soft tissue tumors. Immunohistochemical detection helps identify myogenic differentiation in neoplasms such as leiomyomas, leiomyosarcomas, rhabdomyomas, and rhabdomyosarcomas. However, desmin expression is not exclusive to these tumors, and interpretation generally requires additional markers and morphological assessment. Clone D33 has an extensive history of use in immunohistochemical studies of muscle tissue and tumors, making its established identity an important distinguishing feature. The Muscle-Specific Intermediate Filament Antibody supports research examining muscle lineage and tumor differentiation.
Mutations in the DES gene are associated with desmin-related myopathies and cardiomyopathies, disorders characterized by abnormalities in intermediate filament organization and muscle function. Research into desmin contributes to understanding cytoskeletal stability, organelle positioning, and the mechanical resilience of muscle cells. NSJ Bioreagents offers clone D33 for investigations of desmin expression in muscle biology and pathology. A Desmin Antibody D33 provides a research tool for examining muscle-specific intermediate filaments, cellular differentiation, and the structural organization of muscle tissue.
This antibody can be compared with our Desmin Antibody (clone DES/2960R) for detection of desmin as a muscle marker with validation supported by gene knockdown.
Optimal dilution of the Desmin Antibody D33 / Muscle-Specific Intermediate Filament Antibody should be determined by the researcher.
Human leiomyoma proteins were used as the immunogen for the Desmin Antibody D33.
Store the Desmin Antibody D33 at 2-8oC.
